拉米普利酸对氧化型低密度脂蛋白损伤血管内皮细胞的保护作用

摘    要:体外培养人脐静脉内皮细胞,观察拉米普利酸对由Cu2 、内皮细胞氧化修饰的LDL损伤内皮细胞的保护作用及与NO、cGMP的关系。结果发现:OLDL损伤内皮细胞,使MDA升高,NO、cGMP下降。拉米普利酸能有效对抗OLDL损伤内皮细胞的作用,降低MDA,升高NO、cGMP。一氧化氮合成酶拮抗剂硝基左旋精氨酸(N-L-Arg)能加强OLDL的损伤效应,且能部分拮抗拉米普利酸的保护作用。结果提示:拉米普利酸对OLDL损伤血管内皮细胞的保护作用与其促进内皮细胞NO合成和释放有关。

Abstract:In this study we investigated the effects of ramiprilat on nitritc oxide (NO), cGMP, malondialdehyde(MDA) induced by oxidative modification of low density lipoprotein in cultured human umbilical vein endothelial cells. The results demonstrated that MAD increased significantly,but NO and cGMP decreased significantly wheh endothelial cells stinlulated by OLDL. Nitro-LArginine, a compeptive inhibitor of NO synthase, increased the influensed NO and cGMP of ECstimulated by OLDL. But N-L-Arg could abolish the protective effects of ramiprilat partly. Theseresults suggest that the protective cffects of ramiprilat of endothelial cells induced by OLDL maybe due to stimulation of nitric oxide production or relcarse of the cndothelial cells.
