摘    要:目的:观察虎杖对湿热型高脂血症大鼠病证结合模型的作用及可能的机制。方法:采用高脂饲料饲喂建立基础高脂血症模型,并将其分为模型对照组(高脂饮食+湿热环境组)、模型组(高脂饮食+湿热环境+白酒组)、虎杖(高、低剂量)(15、7.5 g·kg-1)组、反证治疗(干姜,10 g·kg-1)组、血脂康(0.3 g·kg-1)组、辛伐他汀(7.5 mg·kg-1)组,分别给予相应的药物,另设对照组。末次给药后采集标本,检测相关指标。结果:复合因素建立湿热型高脂血症大鼠病证结合模型制备成功。与模型组比较,虎杖高、低剂量组及血脂康组体质量、肝质量及总胆固醇(total cholesterol,TC)、三酰甘油(triglyceride,TG)、低密度脂蛋白胆固醇(low density lipoprotein cholesterol,LDL-C)、白细胞介素-6(interleukin-6,IL-6)、肿瘤坏死因子-α(tumor necrosis factor-α,TNF-α)水平显著降低(P<0.01);肝脏指数显著降低(P<0.05);虎杖高、低剂量组血清三磷酸腺苷结合盒转运体A1(ATP binding casstte transporter,ABCA1)、卵磷脂胆固醇乙酰转移酶(lecithin cholesterol acyltransferase,LCAT)均明显升高(P<0.05),其中虎杖低剂量组作用更显著;而反证治疗组的TC和LDL-C则升高,证明了反证治疗的无效性,在一定程度上佐证了湿热型高脂血症大鼠模型造模成功。结论:虎杖治疗湿热型高脂血症的机制可能与其调节血脂、抑制炎性因子水平及调控胆固醇逆转运通路有关。

Abstract:Objective:To observe the effect of Huzhang(Rhizoma Polygoni Cuspidati)on the combination of disease and syndrome model of hyperlipidemia rats with damp-heat type and its possible mechanism.Methods:The model of hyperlipidemia was established by feeding high-fat diet and divided into model control group(high fat diet+damp-heat environment group),model group(high fat diet+damp-heat environmen+wine),Huzhang(Rhizoma Polygoni Cuspidati)(low and high dosage)(7.5,15 g·kg-1)group,anti-treatment(dry ginger,10 g·kg-1)group,Xuezhikang(0.3 g·kg-1)group,simvastatin(dried ginger,7.5 mg·kg-1).Each group was given corresponding drug treatment.The control group was set up.After the last administration,specimens were collected and indicators were tested.Results:The combined factors established the damp-heat hyperlipidemia rat model combining disease and syndrome successfully.Compared with the model group,the body weight,liver weight,total cholesterol(TC),triglyceride(TG),low density lipoprotein cholesterol(LDL-C),interleukin-6(IL-6),tumor necrosis factor-α(TNF-α)in the high and low dosage groups of Huzhang(Rhizoma Polygoni Cuspidati)and Xuezhikang group decreased significantly(P<0.01);the liver index decreased significantly(P<0.05);serum ATP binding cassette transporter A1(ABCA1)and lecithin cholesterol acyltransferase(LCAT)were significantly increased in the high and low dosage groups of Huzhang(Rhizoma Polygoni Cuspidati)(P<0.05),among which the effect of the low dosage of Huzhang(Rhizoma Polygoni Cuspidati)is more significant;the TC and LDL-C of the counter-syndrome treatment group increased,which proved the ineffectiveness of the anti-treatment,and to a certain extent supported the success of the damp-heat hyperlipidemia rat model.Conclusion:The mechanism of Huzhang(Rhizoma Polygoni Cuspidati)in the treatment of damp-heat hyperlipidemia may be related to its regulation of blood lipids,inhibition of inflammatory factors,and regulation of cholesterol reverse transport pathways.
