HDAC3、IL-17RA、miR-19a-3p在类风湿性关节炎相关间质性肺病小鼠中的表达及意义

摘    要:目的研究HDAC3、IL-17RA、miR-19a-3p在酵母多糖诱导的类风湿性关节炎相关间质性肺病(RA-ILD)小鼠中的表达情况。方法采用注射酵母多糖的方式构建RA-ILD小鼠模型,通过关节肿胀评分及肺脏组织病理评估造模是否成功;测定肺脏羟脯氨酸(HYP)的含量验证模型的可靠性;测定肺组织和肺泡灌洗液中HDAC3、IL-17RA和miR-19a-3p的表达情况。结果 12周后,RA-ILD组小鼠均患有严重的关节炎且部分小鼠肺组织出现明显炎症,HYP含量高于Normal组(P<0.01)。RA-ILD组小鼠肺组织和肺泡灌洗液中的HDAC3、IL-17RA的mRNA表达量均明显高于Normal组,miR-19a-3p表达量低于Normal组(P<0.05)。RA-ILD组小鼠肺组织和肺泡灌洗液中的HDAC3、IL-17RA蛋白表达量均明显高于Normal组(P<0.05)。结论 HDAC3和IL-17RA在RA-ILD纤维化小鼠模型肺组织中高表达,而miR-19a-3p低表达,推测三者可能参与了RA-ILD的发病。

Abstract:Objective To study the expression of HDAC3,IL-17RA and miR-19a-3p in rheumatoid arthritis-associated interstitial lung disease(RA-ILD)mice induced by yeast polysaccharide.Methods The RA-ILD mice model was established by injecting yeast polysaccharide.The success of the model was evaluated by joint swelling score and lung histopathology.The content of hydroxyproline(HYP)in lung was determined to verify the reliability of the model.The expression of HDAC3,IL-17RA and miR-19a-3p in lung tissue and bronchoalveolar lavage fluid were determined.Results After 12 weeks,the RA-ILD group mice suffered from severe arthritis,and some mice had obvious inflammation in lung tissue.The content of HYP in the RA-ILD group was higher than that in the Normal group(P<0.01).The mRNA expressions of HDAC3 and IL-17RA in lung tissue and bronchoalveolar lavage fluid of the RA-ILD group were significantly higher than those of the Normal group,and the expression of miR-19a-3p was lower than that of the Normal group(P<0.05).The protein expressions of HDAC3 and IL-17RA in lung tissue and bronchoalveolar lavage fluid of the RA-ILD group were significantly higher than those of the Normal group(P<0.05).Conclusion HDAC3 and IL-17RA are highly expressed in the lung tissue of RA-ILD fibrosis mice model,while miR-19a-3p is lowly expressed.It is speculated that three factors may be involved in the pathogenesis of RA-ILD.
