Abstract:Objective To explore the effects of SDF-1 on endothelial progenitor cells(EPCs).Methods The mono nuclear cells were isolated from bone marrow of rats by density gradient centrifugation combined with adherence cells filtration,then the isolated cells were plated on fibronectin coated culture dishes.After 7 days,adherent cells were kept with different concentrations of SDF-1 for 48 hours.EPCs proliferation was determined by MTT test;EPCs migration ability assay was performed by Transwell test;The CXCR4 antibody expression in cultured EPCs was examined by IHC.Results SDF-1 increased the number of EPCs in the dose-depending manner(P<0.01or <0.05) : SDF-1 promoted EPCs proliferation and migration capacity(P<0.01),EPCs presented CXCR4 positive expression after induction by SDF-1 in dose-depending manner.Conclusion SDF-1/CXCR4 biological axis can increase the number of bone mar row derived EPCs and improve their biological characteristics.
