干片对免疫组化染色结果的影响

摘    要:目的研究在免疫组化操作全程中,不同环节干片对免疫组化染色所造成的影响。方法随机抽取本科室30例乳腺癌标本,每例各切4张切片,分为4组,同时进行ER标记的免疫纽化染色:第一组按正常步骤操作;第二组在修复时人为造成干片;第三组在修复后人为造成干片;第四组在滴加一抗后人为进行干片。其余操作完全同第一组。比较四组之间阳性表达和背景强度的差异。结果修复液不足时,淹没部分基本不影响阳性表达,而未淹没部分则全部为阴性,分界明显;但背景依然清晰,未受影响。修复后晾干与正常步骤操作的结果无统计学差异(P〉0.05)。而滴加一抗后如果干片,与正常操作步骤结果比较影响很大(P〈0.01):显色偏弱,表达不均,整张片子显得比较黄而且不太透明,边缘效应也较常见。结论在修复时不能干片.否则会造成假阴性;在冲洗步骤造成的干片,对染色结果无影响;在滴加抗体的各个步骤,必须防止干片.以避免造成假阴性及背景染色。

Abstract:Objective To study the influence of dry slides on immunohistochemical staining in different stages. Methods Thirty samples of breast cancer specimens were randomly selected from our department. Each sample was cut into 4 sections and divided into four groups. ER mark staining were performed in four groups with slice dehydrated in different stages: first group was handied by usual procedure; second group was dehydrated when retrieving; third group was dehydrated after retrieving; fourth group was dehydrated after dropping the primary antiboy. The differences of positive expression and background intensity were compared among these four groups. Results The submerged part did not affect the positive expression when the retrieval liquid was insufficient, the part without retrieval liquid submerging presented negative and clear boundaries. However, the background was clear and unaffected. There is no statistical differ- ence between the group which was dried after retrieval and the group which was handled by usual procedure (P〉0.05). Compared with the usual handling procedure, it was affected obviously if the slice was dried after dropping the primary antibody (P 〈0.01). The staining was faint and uneven. The slice presented yellow, opaque and edge-effect was common. Conclusion False negative result is most likely caused if the slice is dried when retrieving. The result of staining is not affected if the slice is dried when rinse. We must prevent dry slides after dropping antibody in order to avoid false negative result and staining background.
