| 摘 要： | 目的建立阿普唑仑血药浓度的反相HPLC测定方法.方法固定相为ODS 色谱柱(粒径5 μm,150×3.9 mm ID);甲醇-0.02M pH=5.0的醋酸铵(60∶40)为流动相;流速为1.0 ml/min;检测波长为223 nm(阿普唑仑的最大吸收波长).结果本法最低检出浓度为5 ng/ml,线性范围为0.01～50 μg/ml,平均回收率为98.67%,日内和日间平均变异系数分别为2.44%和2.74%.结论用HPLC法测定血清中ALP的含量既简便又可行,可作为阿普唑仑血药浓度的测定方法.

| Abstract: | Objective: To determine the alprazolam in serum by RP-HPLC. Methods: Chromatographic conditions were adopted with ODS Column as solid phase, Methanol:NH4Ac=60:40 as mobile phase, the flow rate of mobile phrase at 1.0 ml/min and alprazolam detection at 223nm. Results: The average recoveries of alprazolam were 98.67%(n=5). RSD with in-day was 2.44% and RSD between-day was 2.74%. Conclusion: The method is simple, fast and reliable, and can be used for the determination of ALP in serum. |
