免疫抑制法制备单克隆抗体的研究

用马的红细胞作抗原，先用不含靶抗原的Ｍ２６红细胞和环磷酰胺免疫小鼠，而后再用含靶抗原的Ｍ１７红细胞免疫小鼠。结果显示：经用环磷酰胺抑制后，试验组产生抗非靶抗原抗体的孔数为１５孔，而对照组为１７９孔。试验组获得仅与Ｍ１７红细胞起反应的抗体为１１孔，对照组为８孔。经克隆后，结果显示：在与５５匹含靶抗原的马红细胞反应中。试验组与５２匹起反应，而对照组与４６匹起反应。在与１０１匹不含靶抗原的马红细胞反应中，试验组与３４匹起反应，而对照组与６０匹起反应。这表明，采用环磷酰胺做抑制剂的免疫抑制法，可有效的抑制抗非靶抗原抗体的产生，减少筛选的工作量，而获得的单克隆抗体（单抗）特异性要比对照组强。

STUDY ON MON0CL ONAL ANTIBODIES PRODUCTED BY IM-MUNOSUPPRESSION METHOD

he results showed that there were 179 wells secreting antbodies against non-target anti-gens in the control and l5 wells in the test,there were 11 wells producing antibodies against tar-get antigens in the test aiid 8 wells in the control.After cloning, the resuIts showed that 52 of55 horse red cells which contain target antigens reacted with the monoclonal antibodies(McAb)in the test,and 46 of 55 horse red cells in the control.34 of 101 horse red cells which did notcontain the target antigens reacted with the McAb in the test and 60 of 101 horse red cells in thecontrol.It indicated that cyclophosphamide was successful in suppressing the production of theantibodies against non-target antigens. Therefore it was easy to selecte the McAb concerned.The McAb produced in the test were also more specific than that in the control.
