阿托伐他汀对兔动脉粥样硬化一级预防的实验研究

摘    要:目的:探讨阿托伐他汀对兔动脉粥样硬化的一级预防作用。方法:建立兔动脉粥样硬化模型并随机分成3组,各组兔分别于实验开始前1d及实验开始后的第4、8、12周末采血,提取血清应用日立7170型全自动生化分析仪测甘油三酯(TG)、总胆固醇(TC)、低密度脂蛋白胆固醇(LDL-c)、高密度脂蛋白胆固醇(HDL-c),按试剂盒说明采用不同的方法分别测定高脂和正常血脂标本丙二醛(MDA)后处死动物,留取主动脉作形态学观察以及扫描、透射电镜观察。结果:阿托伐他汀可明显降低血清中TG、TC、LDL-c及MDA(P<0.05),升高HDL-c(P<0.05);阿托伐他汀可明显减轻兔主动脉动脉粥样硬化的产生。结论:阿托伐他汀具有调节血脂、抗氧化作用,可预防兔动脉粥样硬化的发生。

Abstract:Objective: To probe the primary preventive effect of atrovastatin on atherosclerosis in abbits. Methods: Twenty-four male Newzeland rabbits were randomized into 3 groups: control group, cholesterol group and atrovastatin group. The blood samples were drawn from the rabbits just before the experiment and at the fourth, the eighth and the twelfth weekend of the experiment, then they were analysed the total cholesterol (TC), triglyceride (TG), high density lipoprotein cholesterol(HDL-c),low density lipoprotein cholesterol(LDL-c) by 7170 type biochemical auto-analysis apparatus and malondialdehyde (MDA) by different methods in different group. All the rabbits were sacrificed at the twelfth weekend and excised aorta for morphological observation by optical microscope and electron microscope. Results: Compared with the cholesterol group, the serum TC, LDL-c, TG and MDA were appreciably decreased(P<0.05), but the level of HDL-c were increased (P<0.05), and the atherosclerosis were obviously lightened by dietary atrovastatin. Conclusion: Atrovastatin has the efficacy of anti-oxidation and modulating lipoprotein,it can prevent the development of atherosclerosis.
