荧光定量检测乙型肝炎病毒脱氧核糖核酸与乙型肝炎血清标志物的关系研究

摘    要:目的 了解乙型肝炎患者体内乙型肝炎病毒 (HBV)复制及传染性。方法 运用酶联免疫吸附测定 (EL ISA)法检测 HBV血清标志物和荧光定量聚合酶链反应 (FQ- PCR)检测 HBV DNA,并对其结果进行分析比较。结果 乙型肝炎大三阳患者血清 HBV DNA检出率为 10 0 % ,平均拷贝数为 2 .0 9× 10 7/m L,HBV DN A的对数值为 7.3± 1.6 ;小三阳患者血清 HBV DN A检出率为 6 4 .5 % ,平均拷贝数为 6 .76× 10 4 /m L,HBV DNA的对数值为 4 .8± 1.2。结论 同时进行乙型肝炎血清标志物的 EL ISA检测及 HBV DNA的 FQ- PCR检测 ,有利于临床对 HBV感染的诊断、治疗方案的选择及疗效判定。

Abstract:Objective To study the HBV replication and infectivity in patients with hepatitis B.Methods All samples were tested for HBsAg,anti HBs,HBeAg,anti HBe,and anti HBc by ELISA.To investigate the HBV DNA,fluorescence quantitative PCR(FQ PCR) was also performed on all samples.Results In 357 cases with HBsAg(+),HBeAg(+),anti HBc(+) samples,FQ PCR results were positive and the average load of HBV DNA was 2.09×10 7 copies/mL,with a positive rate of 100%.In 207 of 321 cases with HBsAg(+),anti HBe(+),and anti HBc(+) samples,the average HBV DNA load was 6.76×10 4 copies/mL,with a positive rate of 64.5%.Conclusion Both detection methods of serological markers by ELISA and of HBV DNA by FQ PCR are good monitors for the state of HBV infection.The two methods used together can facilitate the approaches to diagnosis,treatment,and assessment of curative effect.
