罗勒多糖对紫杉醇抑制肺腺癌A549细胞增殖作用的影响

摘    要:目的:探讨罗勒多糖对紫杉醇体外抑制人肺腺癌A549细胞增殖作用的影响。方法:采用MTT法测定罗勒多糖对紫杉醇抑制A549细胞增殖作用的影响,倒置相差显微镜下观察罗勒多糖联合紫杉醇导致的A549细胞形态学变化,并采用Hoechst 33342/PI荧光双染法分析罗勒多糖与紫杉醇联合应用所引起的A549细胞死亡方式。结果 :MTT检测和形态学观察表明罗勒多糖对紫杉醇抑制A549细胞增殖具有明显的增效作用,其增效作用呈浓度和时间依赖性;Hoechst 33342/PI检测显示紫杉醇及紫杉醇联合罗勒多糖作用48 h后可见凋亡细胞。结论:罗勒多糖能增强紫杉醇对人肺腺癌A549细胞株增殖的抑制作用,具有化疗增效作用,这种增效作用以诱导细胞凋亡为主要方式,呈浓度和时间依赖性。

Effect of Basil Polysaccharide on Paclitaxel's Function of Inhibiting Proliferation of Lung Adenocarcinoma A549 Cell

Abstract:Objective:To study the effect of basil polysaccharide on paclitaxel's function of inhibiting the proliferation of lung adenocarcinoma A549 cell. Methods:The effect of basil polysaccharide on paclitaxel's function of inhibiting the proliferation of lung adenocarcinoma A549 cell was determined by MTT assay and the morphological changes of A549 cell caused basil polysaccharide combined with paclitaxel were observed by inverted phase contrast microscope. The death manner of A549 cell caused by basil polysaccharide combined with paclitaxel was analyzed by Hoechst 33342/PI double staining method. Results:The MTT results and morphological observation showed that basil polysaccharide had significant synergistic effect on paclitaxel's function of inhibiting the proliferation of A549 cell and the synergistic effect was concentration and time dependent. The Hoechst 33342/PI test showed apoptotic cells 48 hours after the work of paclitaxel and paclitaxel combined with basil polysaccharide. Conclusion:Basil polysaccharide can enhance the inhibitory effect of paclitaxel on the proliferation of human lung adenocarcinoma A549 cell,which means it can enhance the chemotherapeutic effect. Inducing cell apoptosisthe is the main mechanism of the synergistic effect and the synergistic effect is concentration and time dependent.
