白僵蚕体外抗病毒有效部位的筛选

目的:探索中药白僵蚕通过大孔吸附树脂洗脱不同部位的体外抗病毒作用,并进一步分离纯化,为分离抗病毒单体物质做准备。方法:先用不同极性的溶剂提取,观察其抗病毒效果。再用大孔吸附树脂的方法,以蒸馏水、25%的乙醇、50%的乙醇、75%的乙醇洗脱白僵蚕醇提水沉的上清液,收集洗脱液。分别采用EV-71-RD细胞和RSV、HSV-1-Vero细胞体外病毒感染模型,进行体外抗病毒作用评价,通过显微镜观察细胞病变效应(CPE),以酶标仪测定中性红染色A540值,计算治疗指数(TI)。结果:白僵蚕醇提水沉的上清液中水洗脱部位对RSV病毒有一定的抑制作用。结论:白僵蚕抑制RSV病毒的有效部位是醇提水沉后的上清液中水洗脱部位。

To Determine the Effective in Vitro Antiviral Part of Bombyx Batryticatu

Objective:To explore the in vitro antiviral part of Bombyx batryticatu in macroporous adsorption resin eluent,and after further purification to isolate the antiviral monomer. Methods:After extracted with solvent of different polarity,the antiviral effect was observed. Using macroporous adsorption resin method,the supernate of alcohol extraction-water precipitation of Bombyx batryticatu was eluted with distilled water,25%ethanol,50% ethanol and 75% ethanol,and the eluent was collected. The EV-71-RD cells,RSV,HSV-1-Vero cell transfected with virus to evaluate antiviral effect,and cytopathic effect(CPE) was observed under microscope. The neutral red staining A540 value was detected with microplate reader to calculate the therapeutic index(TI). Results:The supernate of the product of alcohol extraction-water precipitation of Bombyx batryticatu could suppress respiratory syncytial virus(RSV). Conclusion:The effect part of Bombyx batryticatu in hibiting RSV was in the supernate of the alcohol extraction-water precipitation
