顺铂致大鼠急性肾功能衰竭时肾皮质脂质过氧化的变化

摘    要:目的 观察不同剂量顺铂致大鼠急性肾衰时肾皮质脂质过氧化的变化 ,探讨顺铂肾毒性的发生机制。方法 雌性Wistar大鼠随机分为生理盐水组、顺铂Ⅰ组、顺铂Ⅱ组、顺铂Ⅲ组 ,均为尾静脉注射给药 ,每d 1次 ,连续 5d。第 6d取血测肌酐 (Scr)、尿素氮 (BUN)、肾皮质组织丙二醛 (MDA)含量、超氧化物歧化酶 (SOD)活性、谷胱甘肽过氧化物酶 (GSH -Px)活性。结果 顺铂组血清Scr、BUN明显升高 ,肾皮质组织MDA含量升高 ,SOD与GSH -Px活性降低 ,与对照组相比均有显著差异 (P <0 0 5 ) ,且肾皮质组织SOD活性、GSH -Px活性与血清Scr、BUN含量呈明显负相关 (P <0 0 5 ) ,肾皮质组织MDA含量与血清Scr、BUN含量呈明显正相关 (P <0 0 5 )。结论 顺铂引起急性肾功能衰竭与肾皮质脂质过氧化增强有关 ,且随剂量增加肾皮质损伤加重

Abstract:Objective To observe the changes of the renocortical tissue lipid peroxicition of acute renal failure induced by diffent dose cisplatin in rats and to study the mechanism of cisplatin nephrotoxicity.Methods Female Wistar rats were randomly divided into NS group,CDDP 1 group,CDDP 2 group, CDDP 3 group.All rats were injected via the tail vein with cisplatin and NS by qd.The changes of Scr,BUN,MDA,SOD,GSH-Px were measured.Results The contents of serum Scr and BUN of CDDP groups were significantly highter than that of NS group ( P <0 05).The content of renocortical tissue MDA was significantly highter than that of NS group ( P <0 05).The activities of renocortical tissue SOD and GSH-Px were lower than that of NS group ( P <0 05).The activities of renocortical tissue SOD and GSH-Px,the contents of serum Scr and BUN were significantly correlative.Conclusions The renocortical tissue lipid peroxiciton increasing might be the cause of cisplatin nephrotoxicity.The renocortical tissue injury becomes heavier by dose increasing.
