大豆异黄酮联合叶酸对胎鼠脑细胞Pax3和Cx43表达的影响

摘    要:目的观察大豆异黄酮联合叶酸对胎鼠脑细胞Pax3和Cx43表达的调控作用及其与神经管畸形发生的关系,并分析其可能的机制。方法将72只孕鼠随机分为9组对照组、模型组(环磷酰胺,CP组)、叶酸干预组(FA)、大豆异黄酮不同剂量干预组(SIF1、SIF2和SIF3)以及与FA配伍的大豆异黄酮联合干预组(SIF1+FA、SIF2+FA和SIF3+FA)。SP免疫组化法检测胎鼠脑神经细胞Pax3及Cx43蛋白的表达,并观察胎鼠外观,记录胎鼠总数和畸胎数。结果(1)与模型组比较,各干预组均能降低NTDs发生率,其中以FA+SIF2联合干预组效果最好;(2)与对照组比较,模型组中Pax3表达下调,Cx43表达过度;与模型组相比,各干预组均有Pax3表达上调,而Cx43表达下调的改变。结论大豆异黄酮、叶酸或两者联合与NTDs形成相关基因Cx43的下调和Pax3的上调有关;Cx43的过度表达、Pax表达的下调可能是CP诱发NTDs重要机制之一。

Abstract:Objective: To investigate the effect of soybean isoflavone(SI) combining with folic acid (FA) on the expression of regulated genes in neurons of rat embryos with neural tube defects (NTDs) induced by cyclophosphamide and its mechanism.Method: Seventy two pregnant rats were randomly divided into control group(C), model group (M), SI intervention groups and SI-FA intervention groups. Pax3 and Cx43 were measured by immunohistochemical method. Results: (1) Compared with M group, the incidence of NTDs in each intervention group was decreased, the lowest in the (SI 160 mg/kg bw FA 0.7mg/kg bw) group; (2) Compared with C group, the expression of Pax3 and Cx43 in M group was significantly decreased and increased respectively (P<0.05). Compared with M group, the expression of Cx43 and Pax3 in intervention groups was significantly decreased and increased respectively. Conclusion: Increased expression of Pax3 and decreased expression of Cx43 might be involved in the protective mechanism of soybean isoflavone and folate on rats NTDs.
