芎芷地龙汤对偏头痛模型三叉神经脊束核c-fos和β-EP蛋白共存表达的影响

摘    要:目的研究芎芷地龙汤对偏头痛模型三叉神经脊束核c-fos和β-EP蛋白共存表达的影响。方法健康雄性SD大鼠随机为4组:生理盐水组、模型组、舒马普坦组、中药治疗组。皮下注射NTG注射剂复制实验性偏头痛动物模型,经药物干预后在4h、6h两个时间点取材,采用免疫组化双标染色法测定三叉神经脊束核c-fos和β-EP的免疫阳性神经元的兴奋状态及其共存表达情况。结果模型组c-fos阳性细胞数增多,两个时间点的阳性细胞数与生理盐水组比较有显著性差异(P0.01);模型组β-EP阳性细胞数4h增多,6h减少,两个时间点的阳性细胞数与生理盐水组比较没有显著性差异。中药治疗组和舒马普坦组c-fos阳性细胞数减少,两个时间点的阳性细胞数与模型组比较有显著性差异(P0.01),中药治疗组β-EP阳性细胞数增多,两个时间点的阳性细胞数与模型组比较有显著性差异(P0.05,P0.01);舒马普坦组β-EP阳性细胞数增多,两个时间点的阳性细胞数与模型组比较有显著性差异(P0.01)。结论芎芷地龙汤可以减少由硝酸甘油诱导的致痛物质c-fos的表达,增加抗痛物质β-EP的表达,从而进一步减少三叉神经脊束核神经元的兴奋性,阻止痛觉信息的传递。

Abstract:Objective:To study the effect of Xiong Zhi Di Long Decoction on coexistence express of c-fos and β-EP in spinal nucleus of trigeminal nerve of migraine model. Methods Healthy male SD rats were randomly divided into 4 groups: normal sodium control group,model control group,Sumatriptan control group,Xiong Zhi Di Long Decoction group. Reproduce migraine animal model with subcutaneous injection NTG,neuronal emotional state and coexistence express of c-fos and β-EP which located at spinal nucleus of trigeminal nerve were determined with immunohistochemical double staining techniques after medicine interfered 4h and 6h.Results:Positive neurons of c-fos of model group were increased,which were significant compared with normal sodium group at 4h and 6h(P0.01). Positive neurons of β-EP of model group were increased at 4h,decreased at 6h,which were not significant compared with normal sodium group at 4h and 6h. Positive neurons of c-fos of Chinese medica group and Sumatriptan group were decreased,which were significant compared with model group at 4h and 6h(P0.01). Positive neurons of β-EP of Chinese materia medica group were increased,which were significant compared with model group at 4h and 6h(P0.05,P0.01).Positive neurons of β-EP of Sumatriptan group were increased,which were significant compared with model group at 4h and 6h(P0.01).Conclusion:Xiong Zhi Di Long Decoction can reduce the expression of c-fos which was induced by NTG,and increase the expression of anti-pain substance β-EP.This effect can depress neuronal excitability in spinal nucleus of trigeminal nerve,and can inhibit nociceptive pain information transmitting.
