线粒体DNA 5178 A/C多态性与2型糖尿病肾病相关性

目的探讨线粒体DNA 5178A／C多态性与2型糖尿病肾病（DN）的相关性。方法将1039例2型糖尿病患者分为无糖尿病肾病（DN-0）组和糖尿病肾病（DN）组，后者进一步分为微量蛋白尿（DN-1）组和大量蛋白尿（DN-2）组，进行病例对照研究。采用聚合酶链反应-限制性片段（PCR-RFLP）技术检测线粒体DNA5178A／C多态。结果（1）2型糖尿病者尿白蛋白肌酐比值（ACR）与5178A／C多态无相关。（2）DN-0，DN-1及DN-2这3组间基因型频率的差异无显著性。结论线粒体DNA 5178A／C多态性与中国2型糖尿病患者ACR水平无相关，与糖尿病肾病无相关。

Association of the mitochondrial DNA 5178 A/C polymorphism with diabetic nephropathy in Chinese type 2 diabetic patients

Purpose To observe the relationship between diabetic nephropathy(DN) and the mitochondrial DNA 5178 adenine/cytosine(mt 5178 A/C) polymorphism in Chinese type 2 diabetic patients. Methods A case-control study for a total of 1039 Chinese type 2 diabetic patients including 524 patients without diabetic nephropathy and 515 patients with diabetic nephropathy for controls was performed.The patients with diabetic nephropathy(DN;n=515)were further subdivided into microalbuminuric patients(DN1;n = 430) and macroalbuminuric patients(DN2;n =85).The mt 5178 A/C was genotyped by the polymerase chain reaction-restriction fragment length polymorphism method(PCR-RFLP). Results(1) The Mt 5178 C genotype was not associated with the albumin-to-creatinine ratio(ACR).(2) No significant difference in genotype distribution was observed among the DN0,DN1 and DN2groups. Conclusions The Mt 5178 C/A polymorphism was not associated with the ACR and DN among Chinese type 2 diabetic patients.
