紫穗槐种子体外抗肿瘤活性部位的筛选

摘    要:目的 筛选紫穗槐种子体外抗肿瘤的活性部位.方法 利用溶剂法从紫穗槐种子中提取总有机成分,利用乙醇-水梯度洗脱的D101大孔树脂柱层析法获得各极性部位,利用高效液相色谱分析方法检测各部位的主要成分,利用噻唑蓝(MTT)法评价各部位的体外抗肿瘤活性.结果 D101大孔树脂柱层析获得的各部位高效液相色谱色谱峰差异明显.90%乙醇洗脱(F90)部位具有显著的细胞毒性,用药72 h时对A549、HepG2、MCF-7、HCT-116的IC50值分别为8.58±0.59、5.57±0.44、6.94±0.52、9.03+0.63 μg·mL-1,其它各部位的细胞毒性均不明显.化学成分指认表明,F90中的主要成分为鱼藤酮类似物.结论 D101大孔树脂柱层析法可以有效的将紫穗槐种子中的化学成分进行极性分段,90%乙醇洗脱部位为抗肿瘤活性部位,主要活性成分为鱼藤酮类成分,但其确切的化学成分和作用机制有待进一步的研究.
