摘    要:观察药用猪胆酸钠诱导体外培养的人早幼粒白血病细胞系HL-60细胞分化过程中超微结构的变化及其细胞呼吸爆发功能特征.猪胆酸钠(400mg·L-1终浓度)诱导HL-60细胞分化过程中,细胞超微结构发生明显改变,细胞核浆比例降低,细胞核变小,核仁减少,胞核出现深浅不等的凹陷;多数细胞出现不规则的表面突起,细胞表面伸出不规则大型钝形伪足,细胞浆内可见部分线粒体空泡化及内质网扩张,核内染色质凝聚成块并边集,核扭曲,断裂.同时,用电子自旋共振波谱仪直接检测到猪胆酸钠诱导分化的HL-60细胞发生呼吸爆发时产生羟自由基,并消耗细胞外介质中的氧

Changes in ultrastructure and respiratory burst function of HL 60 cells during differentiation induced by swine bile acids sodium salt
Abstract:The changes in ultrastructure of HL 60 cells during differentiation induced by swine bile acids sodium salt (SBANa) in in vitro liquid cultures were studied by electron microscopy. After cultured with SBANa at 400 mg·L 1 for 2-9 d, HL 60 cells differentiated along myeloid pathway, and the ultrastructure revealed remarkable changes: the proportion of nucleus to cytoplasm decreased, the nuclei became smaller and indentation or condensation, nucleoli reduced, the chromatin clotted or aggregated; most of the cells appeared large blunt pseudopods on the cellular surfaces, and the cells became star like; some of mitochondria exhibited vesiculated and parts of endoplasmic reticulum enlarged in the cytoplasm. By means of electron spin resonance method, it was demonstrated that hydroxyl radicals were generated and more oxygen in the culture medium was taken up by the HL 60 cells induced by SBANa at 400 mg·L 1 for 3 d during the respiratory burst course.
