成骨生长肽促进大鼠骨量增加的作用

摘    要:探讨成骨生长肽(OGP)对骨量增加的作用。方法采用低钙饲料饲养成为骨质疏松模型的50只SD大鼠,随机分为5组,每组10只,分别皮下隔日注射,A组(低剂量OGP)、B组(高剂量OGP)、C组(人降钙素)、D组(鲑鱼降钙素)和E组(生理盐水)对照,共12周。测定血清碱性磷酸酶(ALP)、骨钙素(BGP)、骨密度(BMD),扫描电镜和透射电镜观察。结果OGP治疗组碱性磷酸酶、骨钙素和骨密度明显增高(P<0.01),扫描电镜观察到A、B组骨小梁粗大、完整、间隙小,表面光滑,对照组骨小梁变细、断裂,空隙大。透射电镜观察到A、B组成骨相骨细胞较多,E组退变相骨细胞较多。结论OGP能刺激成骨细胞的活性,能提高骨量,有可能成为治疗骨质疏松症的有效药物

Experimental Study of Osteogenic Growth Peptide Promoting Bone Mass in Rats' Osteoporosis
Abstract:Purpose To study the osteogenic growth peptide(OGP) promoting bone mass in tats' osteoporosis. Methods Fifty male SD rats about 240 g weight and 12 weeks old had been fed by low calcium diet (Ca: 0.1% ,P: 0.6%) for six months.Then osteoporosis model had been done and the rats were allocated randomly into 5 groups,one group used as a control and the rest groups were treated respectively with OGP(low dosage),OGP(high dosage),CT(human calcitonin), CT(salmon calcitonin).The experimental method was that subcutaneous injection and one time every two days for three months.The parameter to be assayed after treatment were serum ALP, BGP,histomorphometry,bone density and shape of bony trabecula. Results The results showed that the level of serum ALP,BGP and the bone density and shape of bony trabecula in two OGP groups were all significantly increased ( P <0.01) compared with the control group.The scan electronic microscope (SEM) showed that the bone trabecula of the therapy groups were thick,continuous and that of the control group was thin, discontinuous.The transmission electronic microscope (TEM) showed that the osteoblast of the therapy groups developed osteogenic phase and the osteoblast of the control group developed degenerative phase. Conclusions The OGP could stimulate osteoblast activity and promote bone mass effect was as well as CT,so OGP have a potential role in osteoporosis.
