摘 要: | 将由SV40病毒晚期启幼子调控的戊型肝炎病毒第三开放读码框架(ORF3)cDNA质粒pSVL-HEVORF3直接注射到小鼠骨骼肌肉,引发了小鼠抗-HEV抗体反应,为HEV基因疫苗的研制提供了可能性。

Abstract: | Gene vaccine,or antigen-expression DNA for elicitation of a specific immune response, represents s new approach to immunization. In this study, a plasmid containing hepatitis E virus (HEV) genome ORF3, driven by SV40 late promoter,has been tested for the ability to elicite the antibody response against HEV in mice. After 1-2 direct injections of 100 ug plasmid DNA into skeletal muscles of mice hind legs, all 3 mice inoculated were positive for anti-HEV IgG in sera by enzyme-linked immunoassay (ELISA). The highest anti-HEV titer reached 1: 20 dilution. This study indicates that the direct inoculation of the plasmid DNA containing HEV cDNA into mice muscles is able to elicite a specific antibody response aganist HEV. |
