摘    要:目的研究知母有效成分(化合物9714)对拟血管性痴呆(VD)模型大鼠学习记忆的影响及其作用机制。方法采用双侧颈总动脉结扎(Commoncarotidarteryocclusion,CCAO)致拟VD模型大鼠,连续给药40d,以Morris水迷宫检测大鼠学习记忆能力,以免疫组化法检测大鼠脑皮质BDNF、ICAM1、VCAM1的表达。结果化合物971410、20、40mg·kg-1组能明显缩短定位航行试验中d2、d3的逃避潜伏期,提高空间搜索试验中原平台象限游泳距离占总距离的百分比(P<0.05,P<0.01);20、40mg·kg-1组能明显增加模型动物脑皮质BDNF的表达,降低脑皮质ICAM1的表达(P<0.05,P<0.01)。结论化合物9714能提高拟VD大鼠的学习记忆能力,同时对缺血后的神经元损伤、炎性损伤具有一定的保护作用。

Abstract:Aim To study the effects and mechanism of effective component in Zhimu (compound 9714) on ability of learning and memory in vascular dementia rats induced by common carotid occlusion. Methods Morris water maze test was used to detect the ability of learning and memory of VD rats. Immunohistochemistry test was used to detect the expression of BDNF, ICAM-1, VCAM-1. Results Compound 9714 10, 20, 40 mg·kg -1 shortened the escape latency time of the second and third days in place navigation test and increased the percent of primary flat floor quadrant swimming distance to the overall swimming distance in spatial probe test in model rats(P<0.05, P<0.01). 20, 40 mg·kg -1 markly inhanced the expression of BDNF and decreased the expression of ICAM-1(P<0.05, P<0.01). Conclusion Compound 9714 had protective effects on VD rats, inhancing the ability of learning and memory, relieving the injury of neuron and inflammatory reaction.
