P[9]轮状病毒受体结合特征及其人群抗体水平与HBGA相关性研究

摘    要:目的 表达P9]轮状病毒(rotaviruses,RV)GST-VP8*蛋白,研究其与组织血型抗原(histo-blood group antigen,HBGA)受体的结合方式。了解广东省汕尾市人群P9]RV特异性抗体水平,并分析其与HBGA受体的相关性。方法 构建pGEX-4T-1- VP8*重组质粒,利用原核表达系统表达纯化VP8*蛋白,目的蛋白经Western bolt确定其特异性,采用酶联免疫吸附试验检测GST-VP8*蛋白与唾液HBGA受体结合方式。利用VP8*蛋白检测广东省汕尾市人群P9]RV抗体,并检测该人群HBGA受体表型,分析P9]RV抗体与HBGA的相关性。结果 成功表达了P9]RV VP8*蛋白,并发现其与A型分泌型HBGA受体特异结合,不结合B、O型分泌型及非分泌型HBGA受体。A型、Le b+/Le y+和分泌型HBGA个体配对的血清中P9]RV特异性IgG阳性率明显较高。结论 P9]RV识别A型分泌型HBGA受体,这将有助于理解RV的流行病学,为RV进一步防控提供试验依据。

Abstract:We express the GST-VP8* proteins of P9] rotavirus and explore its binding pattern with HBGA receptor. We detect P9] rotavirus antibody in Shanwei area of Guangdong, and analyze the association of positive antibody rate with HBGA receptor. The recombinant plasmid pGEX-4T-1- VP8* was conducted and expressed. The binding pattern of P9] RV GST-VP8* proteins and HBGA receptor were identified by enzyme-linked immunosorbent assay(ELISA). The HBGA phenotypes of the individuals in Shanwei of Guangdong were determined in Saliva samples. P9] RV IgG antibodies in sera was measured by ELISA using P9]VP8* proteins as capture antigens to analysis the correlation between P9] RV IgG and HBGA. In this study, 9] RV VP8* proteins were successfully expressed. P9] RV VP8* proteins bind to type A secretor, but not to B、O secretor or all nonsecretor. The correlation between positive rate of P9] specific IgG in serum and host Lewis and secretor phenotypes has been found among 206 studied serum samples, which would help further understanding of rotavirus epidemiology and provides a scientific basis to prevent and control rotavirus.
