H_2O_2预处理抑制α-Synuclein的聚集并减轻Rotenone对PC12细胞的损伤

摘    要:目的 探讨H_2O_2预处理(HPP)对Rotenone诱导PC12细胞损伤及α-Synuclein聚集的影响.方法 采用四甲基偶氮唑盐法(MTF法)、流式细胞仪、免疫印迹(western blot)技术及共聚焦显微镜分别检测细胞活力、细胞凋亡率、α-Synuclein的表达及聚集.结果 H_2O_2预处理能够减少α-Synuclein寡聚体的表达及聚集体的形成,增加PC12细胞活力(Rot组47%±3.67%,HPP+Rot组82%±6.69%),抑制细胞凋亡(Rot组49.68%±4.55%,HPP+Rot组18.73%±5.64%).结论 H_2O_2:预处理能减轻Rotenone对PC12细胞的毒性损伤作用,这种保护作用可能是通过抑制α-Synuclein寡聚体表达实现的.
Abstract:
Objective To explore the effect of hydrogen peroxide preconditioning on the aggregation of α-Synuclein and the injury of PC12 cells induced by rotenone. Methods MTT assay, flow cytometry analysis,western blotting and confocal microscopy were utilized to determine the cell viability, apoptotic ratio of PC12, the expression of α-Synuclein and the formation of α-Synuclein oligomers. Results Hydrogen peroxide preconditioning could down-regulate expression of α-Synuclein oligomers, inhibit the formation of α-Synuclein aggregation,increase cell viability (Rot group 47% ± 3.67% , HPP + Rot group 83.82%±6.69% ) ,and inhibit cell apoptosis ( Rot group 49.68%±4.55% ,HPP + Rot group 18.73%±5.64% ). Conclusion Hydrogen peroxide preconditioning can protect PC12 cells against Rotenone toxicity through downregulating the expression of α-Synuclein oligomers and inhibiting the formation of α-Synuelein aggregation.
