朝医山药补肺元汤对哮喘模型小鼠气道炎症的影响

摘    要:目的:探讨朝医山药补肺元汤对哮喘小鼠气道炎症和Thl/Th2类细胞因子变化的影响及其可能机制。方法:雄性BALB/c小鼠32只随机分成4组。分别采用酶联免疫吸附法(ELISA)和蛋白质印迹检测支气管肺泡灌洗液(BALF)中白细胞介素-4(IL-4)、IL-5、IL-13及γ-干扰素(IFN-γ)含量,观察BALF中炎症细胞和肺组织病理学改变。结果:与正常对照组比较,哮喘模型组小鼠BALF中炎症细胞计数、IL-4、IL-5、IL-13水平和IFN-γ水平显著降低(P0.05)。与哮喘模型组比较,朝医山药补肺元汤低、高剂量组小鼠BALF中炎症细胞计数、IL-4、IL-5、IL-13水平降低,IFN-γ水平显著上升(P0.05)。朝医山药补肺元汤能明显减轻哮喘小鼠肺组织病理学改变,朝医山药补肺元汤低、高剂量组之间比较,无显著差异。结论:朝医山药补肺元汤对哮喘小鼠具有保护作用,其机制可能与调节IL-4/IFN-γ平衡失调、减轻炎症细胞浸润有关。

Abstract:Objective:To explore the protective effects and mechanism of ethanol extract of Shan Yao Bu Fei Yuan Decoction injection on asthmatic mouse.Methods:OVA was injected intraperitoneally and inhaled to produce the asthmatic model.Thirty two mouse were randomly divided into four groups:control group,asthma group and inonotus obliquus groups of high and low dose.The concentrations of IL-4,IL-5,IL-13 and IFN-γ in BALF,were respectively measured by ELISA.The numbers of inflammatory cells in BALF and histropathology changes were observed.Results:In asthmatic group,the number of inflammatory cells and the concentrations of IL-4,IL-5,IL-13 in BALF,but IFN-γ were lower than those in normal control mouse(P<0.05).In Inonotus obliquus group,the number of inflammatory cells,the concentrations of IL-4,IL-5,IL-13 in BALF(P<0.05),and histropathology damage was alleviated significantly.The efficacies in the inonotus obliquus groups of high and low dose were similar,which no significant difference was observed among them.Conclusion:Inonotus obliquus effectively treats asthma by inhibiting the expression of phosphor-p38 MAPK,correcting the inbalance of IFN-γ/IL-4 and decreasing the number of inflammatory cells.
