人脑胶质瘤中EGFR和CTGF的表达及意义

摘    要:【目的】研究表皮生长因子受体(EGFR)和结缔组织生长因子(CTGF)在人脑胶质瘤内的表达,探讨它们与胶质瘤病理分级的关系及其在胶质瘤发生过程中的作用。【方法】采用免疫组织化学SP法,检测64例脑胶质瘤、5例正常脑组织中EGFR和CTGF的表达。用统计学方法分析其表达及与胶质瘤病理分级的关系。【结果】64例胶质瘤患者中EGFR及CTGF阳性表达数分别为41例和43例。正常脑组织均不表达EG—FR和CTGF。EGFR在Ⅰ、Ⅱ、Ⅲ、Ⅳ级胶质瘤中的阳性率分别为12.5%、41.7%、73.9%、85.7%;CTGF在Ⅰ、Ⅱ、Ⅲ、Ⅳ级胶质瘤中的阳性率分别为12.5%、50.0%、78.3%、90.5%。EGFR、CTGF在不同级别胶质瘤间的表达有显著性差异(P〈0.05),两者随着胶质瘤病理分级增高而表达增强(r=0.327,P〈0.01)。【结论】EGFR和CTGF在胶质瘤内均有表达,且随着病理级别的增高而增加,提示EGFR和CTGF在胶质瘤的发生过程中可能起重要的作用,其水平高低与胶质瘤的恶性程度有关。

Abstract:[Objective] To investigate the expression of epidermal growth factor receptor(EGFR) and connective tissue growth factor(CTGF) in human brain glioma, and explore their relationship with pathologic grading and their role in the tumorigenesis of glioma. [Methods]SP immunohistochemical method was used to examine the expression of EGFR and CTGF in 64 human glioma specimens and 5 normal human brain tissue specimens. The relationship between the expression of EGFR and CTGF and pathological grading in human glioma was analyzed by statistical methods. [Results] In 64 cases of glioma, 41 cases had positive expression of EGFR and 43 cases had positive expression of CTGF. EGFR and CTGF were not expressed in normal brain tissue. The positive rate of EGFR in gradeⅠ ,grade Ⅱ,grade Ⅲ ,grade Ⅳ glioma was 12.5%, 41.7%, 73.9% and 85.7%, respectively. The positive rate Of CTGF in grade Ⅰ ,grade Ⅱ,grade Ⅲ ,gradeⅣ glioma was 12. 5 %, 50.0%, 78.3 % and 90.5%, respectively. There was statistically significant difference in the expression of EGFR and CTGF among different grades of glioma(P〈0.05). The expression level of EGFR and CTGF increased with the rise of tumor grade(r=0. 327, P 〈0.01). [Conclusion]EGFR and CTGF are expressed in the gliomas. The level of EGFR and CTGF in gliomas increases with the rise of tumor grade. The results indicate that EGFR arid CTGF may play an important role in the tumorigenesis of glioma, and be related to the malignancy of glioma.
