| 摘 要： | 目的：探讨环氧化酶抑制剂NS398对肾癌786—0细胞生长及bcl-2基因和蛋白表达的影响。方法：用不同浓崩的NS398（0、50、100、150μmol／L）处理肾癌786—0细胞不同时间，采用MTr比色法分析细胞生长抑制率。用不同浓度NS398到理肾癌786—0细胞24h，RT—PCR检测bcl-2mRNA表达的变化，Westernblot检测bcl-2蛋白表达的变化。结果：随着NS398《浓度及处理时间的增加，肾癌786—0细胞生长抑制率升高；随着NS398剂量的增加，bcl-2 mRNA及蛋白的表达水平逐渐减少。结论：NS398可呈剂量和时间依赖性抑制肾癌786—0细胞的生长，呈浓度依赖性减少bcl-2 mRNA及蛋白的表达。NS398可崩通过抑制bcl-2的表达，抑制肾癌786—0细胞的生长。

| Abstract: | Objective To study the effects of NS398 on the expressions of bcl-2 in renal cell carcinoma cells 786-0.Method After 786-0 cells were treated with different concentration NS398(0,50,100,150μmol/L) for 12,24,48 or72h,the growth inhibitory rate of cell was evaluated with MTT assay.The mRNA expressions of bcl-2 were detected by RT - PCR.Protein expressions of bcl - 2 were detected by Western blot Results The cell growth inhibition rate was increased by NS398 with increased of concentration and time in 786-0 cell lines.RT - PCR and Western blot revealed that NS398 decreased the mRNA and protein expressions of bcl -2 with increased of concentration. Conclusion NS398 inhibits 786-0 cell growth in a dose-and time-dependent manners.The expressions of bcl-2 are decreased by NS398 in a dose-dependent manner.NS398 can inhibit proliferation of renal cell carcinoma cells 786-0 in vitro,which may be induced by decreased of the bcl-2 expressions. |
