| 摘 要： | 目的建立测定丁公藤药材中总东莨菪内酯含量的HPLC。方法采用ShimpackCLCODS色谱柱(6mm×150mm,5μm),以甲醇005%磷酸(39∶61)为流动相,检测波长为345nm。结果东莨菪内酯的线性范围为00301～301μg(R2=10000,n=6),检测限为009ng(S/N=3),定量限为03ng(S/N=10),平均回收率为9760%,RSD=159%(n=9)。结论本方法操作简便、准确、灵敏度高,可作为丁公藤药材中总东莨菪内酯的定量分析方法。

| Abstract: | OBJECTIVE To establish a HPLC method for the determination of total scopoletin in Caulis Erycibes.METHODS The separation was accomplished on a Shim-pack CLC-ODS column (6mm×150 mm,5μm) with a mixture of methanol-0.05%H3PO4 (39∶61) as the mobile phase at the flow rate of 1 mL·min-1 . The eluates were detected at 345 nm.RESULTS The linear range for the determination of scopoletin was 0.030 1～3.01μg(R2=1.000 0,n=6),the detection limit was 0.09 ng (S/N=3) and the quantification limit was 0.3 ng(S/N=10).The average recovery was 97.60% and RSD was 1.59%(n=9).CONCLUSION The established method is a simple, accurate and sensitive way to control the quality of Caulis Erycibes. |
