摘    要:目的 观察一氧化碳(CO)暴露对大鼠脑中p53基因的影响。方法 用Wistar大鼠制成CO中毒动物模型,分别在染毒后当天,第2,3天处死,取脑制成切片,用地高辛p53 mRNA探讨原位杂交,观察p53的信号分布。结果 p53 mRNA的表达在海马,大脑皮层和胼胝体比较明显,其中海马的表达信号最强,大脑皮层次之,胼胝体最弱,其他部位呈散在的弱信号。相应区域在各个时间的差异无显著性。对照组未见表达。结论 CO暴露对海马,大脑皮层和胼胝体的损害最为明显;凋亡的过程是一个阶段性持续的过程。

Abstract:Objective To observe the expression of p53 mRNA in brain of Wistar rats after carbon monoxide exposure. Methods The model of CO exposure was made with Wistar rats. Group 1 was control Group 2, group 3 and group 4 were separately stayed in atmospheric condition after CO exposure and then were killed at the third, the second and the first day after the CO exposure respectively. The expressions of p53 gene in the brains were studied using in situ hybridization with p53 mRNA probe. Results The results revealed that the expressions of p53 mRNA were greatly enhanced in hippocampus, cerebral cortex and callosum after the CO exposure. The signal of hybridization in hippocampus was the strongest. There was no marked expression in group 1. The expressions of the gene were not obviously different in groups 2,3,4 and at various time points. Conclusions The main damage by CO exposure occur in hippocampus, cerebral cortex and callosum and apoptosis will remain in the damaged areas for a period of time after the CO exposure.
