摘    要:目的探讨4种灭菌方法对塑料合剂瓶、塑料软膏盒、塑料眼膏管(简称内包装材料)的灭菌效果。方法取未灭菌和经灭菌的内包装材料各10个,分别加入生理盐水溶液荡涤后稀释成供试液,置直径90 mm的无菌平皿中,用营养琼脂培养基、玫瑰红钠培养基培养,检查细菌数、霉菌数;用BL增菌液经增菌、分离培养后,进行革兰染色、生化试验等,检查大肠埃希菌;用营养肉汤、卵黄高盐琼脂平板培养,检查金黄色葡萄球菌;用BL增菌液、溴化十六烷基三甲胺平板培养,检查铜绿假单胞菌。结果环氧乙烷灭菌法灭菌后细菌数(3次平均值,下同)<10个/mL,霉菌数<10个/mL;84消毒液浸泡法灭菌后细菌数13个/mL,霉菌数<10个/mL;紫外线灭菌法灭菌后细菌数381个/mL,霉菌数19个/mL;蒸馏水冲洗烘干法灭菌后细菌数363个/mL,霉菌数18个/mL;4种灭菌法均未检出大肠埃希菌、金黄色葡萄球菌、铜绿假单胞菌。结论环氧乙烷灭菌法灭菌效果最佳,84消毒液浸泡法灭菌效果较好,紫外线灭菌法和蒸馏水冲洗烘干法效果较差。

Abstract:Objective It is to explore the sterilization effects of four kinds of sterilization methods on plastic bottles,plastic eye tube(hereinafter referred to as packaging material).Methods 10 packaging materials not been sterilized and 10 been sterilized were chosen and added with 0.9% normal saline to make test solution after they were oscillated and diluted.Then the solution was poured into the plate whose diameter was 90 mm.Some solution was cultured in nutrient agar and Rose Red sodium medium to detremine the number of bacteria and mold.Some solution was determined by Gram stain and biochemical tests after enrichment with PL and isolated culture to analyze Escherichia coli.Some solution was cultured with high-salt agar plate of nutritional broth and egg to determine staphylococcus aureus.Some was cultured with BL and BLCTMAB plate to analyze Pseudomonas aeruginosa.Results After sterilization by ethylene oxide,the bacterial content and mold content(3times the average,the same below)were both <10/mL.After sterilization by 84disinfectant,the bacterial content and mold content were 13/mL and 10/mL.After sterilization by ultraviolet light disinfection,the bacterial content and mold content were 381/mL and 19/mL.After sterilization by distilled water rinse drying method,the bacterial content and mold content were 363/mL and 18/mL.Escherichia coli, staphylococcus aureus and Pseudomonas aeruginosa weren't found by these four kinds of sterilization methods.Conclusion Sterilization effect of ethylene oxide is the best,the effect of 84 disinfectant is better,that of ultraviolet light disinfection and distilled water rinse drying method were bad.
