“523”药物对受照小鼠造血系统的影响

摘    要:“523”是我所研究的抗放新药,不仅能提高受照小鼠的活存率,对其造血作用也有明显影响。我们研究了“523”药物对受8Gy照射小鼠外周血白细胞的作用,及对内、外源性脾结节和骨髓粒系祖细胞的作用。并对骨髓有核细胞周期作了进一步的研究。结果表明,照前应用“523”可使外周血白细胞明显升高,骨髓和髓外造血指标也明显增加.使正常小鼠骨髓有核细胞由G_0/G_1期向S期转化,从而使S期的细胞比例明显增加.提示“523”对受照小鼠外周血白细胞的作用可能是其作用于骨髓,使骨髓造血细胞增加所引起,而对骨髓的保护作用则是作用于骨髓有核细胞,使骨髓有核细胞处于S期的比例增加有关。

Abstract:523" is a new anti-radiationdrug developed in our institute' Itnot only increases the surviyal ratebut also proatotes he.matopoiesis ofirradiated mice' In this expsriment,'523" was adtniaistered to mice before or aftgr exposttre, and changesin numbers of WBC, endogenous andexogenous colony forming uni t--spleen(CFU--C, CFU--S ) and bone marrowgranular hematopoi et ic progenit orcells were observed' The effect of"523" on the cell cycle of bonemarrow cells was also studied' Theresults showed that when "523" wasgiven before irradiation, the numberof total WBC increased significantly,and CFU--C, CFU--S, CFU--GM alsoincreased significantly. The transitionof Go/GI to S phases in bone marrowcells was promoted, and the cells inS phase increased significantly' Theseresults indicated that "523" may promote the recovery of WBC. whichmight be caused by its protectiveeffect on hetnatopoietic stern cellsthrough its promoting division anddifferentiation and increasing thepercentage of hematopoietic stem cellsin S phase'
