VEGF-C及MMP-12蛋白的表达与非小细胞肺癌纵隔淋巴结转移的相关性研究

摘    要:目的通过检测血管内皮细胞生长因子-C(VEGF-C)及基质金属蛋白酶-12(MMP-12)在非小细胞肺癌(NSCLC)组织及癌旁组织中的表达特点,探讨两种蛋白的表达与NSCLC纵隔淋巴结转移(MLNM)的相关性。方法采集60例NSCLC组织及其癌旁组织,应用免疫组织化学方法检测VEGF-C及MMP-12在两种组织中的表达情况,分析其与临床病理因素之间的关系,以及与纵隔淋巴结转移的相关性。结果 VEGF-C及MMP-12蛋白在NSCLC组织中呈高表达,分别为96.7%(58/60)和91.7%(55/60),显著高于癌旁组织(χ2=63.83、58.85,P<0.01)。同时,二者在有纵隔淋巴结转移的癌组织中的表达水平显著高于无纵隔淋巴结转移的癌组织(χ2=20.317、P=0.000,χ2=5.644、P=0.018)。二者的表达水平与性别、年龄、分化程度以及组织学类型无关(P>0.05),但与术后肿瘤病理学分期(PTNM)有关(P<0.05)。在纵隔有淋巴结转移的癌组织中,VEGF-C与MMP-12蛋白的表达存在相关性(c=0.372,P=0.016)。结论 VEGF-C及MMP-12蛋白在NSCLC组织及癌旁组织中的高表达现象与NSCLC的淋巴结转移密切相关,临床上抑制VEGF-C及MMP-12蛋白的表达有可能为NSCLC的治疗提供新的靶点。

Abstract:Objective Through detecting the expressions of VEGF-C and MMP-12 in the tissues of non-small cell lung cancer(NSCLC) and peritumoral tissues,to investigate their correlation with mediastinal lymph node metastasis of NSCLC.Methods The expressions of VEGF-C and MMP-12 in 60 cases of NSCLC tissues and peritumoral tissues were detected by immunohistochemistry,their correlations with clinical-pathological findings and mediastinal lymph node metastasis of cancer tissues were analysed.Results The positive rates of VEGF-C and MMP-12 were 96.7%(58/60),91.7%(55/60),respectively,which were significantly higher than those of the tissues of peritumoral(χ2=63.83,χ2=58.85,P<0.01).The expression of proteins in mediastinal lymph node metastasis of cancer tissues was significantly higher than that without mediastinal lymph node metastasis of cancer tissues(χ2=20.317,P=0.000;χ2=5.644、P=0.018).The proteins in NSCLC were not correlated with gender,age,degree of differentiation and types of histological(P>0.05),but correlated significantly with the stage of PTNM(P<0.05).Significant correlations between VEGF-C and MMP-12(c=0.372,P=0.016)were observed in mediastinal lymph node metastasis of cancer tissues. Conclusion Close correlation exists between the high expression of VEGF-C and MMP-12 and mediastinal lymph node metastasis in tissues of NSCLC.The proteins may be a new target sites in the treatment of NSCLC.
