硬皮病皮损CTGF,COL-Ⅰ Ⅲ的检测及意义

目的研究硬皮病(SD)发病与结缔组织生长因子(CTGF)及Ⅰ,Ⅲ型胶原(COL-Ⅰ,Ⅲ)表达的关系,探讨硬皮病可能的发病机制。方法采用免疫组化SP法检测72例SD患者皮损中CTGF蛋白及COL-Ⅰ,Ⅲ蛋白表达,18例正常人皮肤组织作为正常对照组。结果①CTGF在SD患者皮损的表达强于对照组,在系统性硬皮病(SSc)表达高于局限性SD(P均<0.001);②COL-Ⅰ在SD患者皮损的表达强于对照组,但在SSc组、局限性SD组间表达无明显差异(P>0.05);COL-Ⅲ在SD患者皮损的表达与对照组相近(P>0.05);相关分析表明COL-Ⅰ与CTGF的表达呈正相关(r5SC=0.728,rSD=0.71,P均<0.01)。结论COL-Ⅰ与CTGF在SD患者皮肤硬化过程中起一定作用,可能与SD的病理纤维化形成有关。

The Expression of Connective Tissue Growth Factor and Collagen Type Ⅰ, Ⅲ in the Scleroderma Skin Lesion

Objective To investigate the role of connective tissue growth factor(CTGF)and collagen typeⅠ(COL-Ⅰ),Ⅲ(COL-Ⅲ)in the pathogenesis of the scleroderma skin lesion.Methods The Expression of CTGF and collagen typeⅠand Ⅲ in the skin lesions from 72 patients with SD and skin from 18 normal controls were detected by immunohistochemical method.Results ①There was a significantly stronger expression of CTGF of the SD group in the lesional skin than that in normal controls(P<0.001,respectively),and significant difference was found between the localized SD and SSc patients.②The expression of collagen typeⅠin the skin lesions of SD group was higher compared with that of the controls(P<0.001),and significant difference was not found between the localized SD and SSc patients.There was no significant difference of collagen TypeⅢamong the localized SD,SSc patients and normal controls(P>0.05 respectively);and the direct correlation between the Expression of CTGF and collagen typeⅠin the SD group of sclerotic skin were found(P<0.05).Conclusion CTGF and collagen typeⅠwere produce intensely in the skin lesions of the SD group,there are direct correlation between the expression of CTGF and collagen typeⅠin the skin lesions of scleroderma patients,this implies that they may be involved in the pathogenesis of scleroderma.
