摘    要:摘要:目的 检测EMA-RTQ-PCR快速鉴别沙门菌死活细胞方法的应用效果,评价该方法的实际应用价值。方法 利用EMA-RTQ-PCR以及国标GB4789.4-2010对人工模拟样品及实际样品分别进行检测。结果 对于人工模拟死菌样品,EMA-RTQ-PCR与国标法检测结果一致,均为阴性;对于人工模拟死活菌混合样品,EMA-RTQ-PCR直接检测结果与国标法存在差异,EMA-RTQ-PCR为阴性,国标法为阳性;人工模拟死活菌混合样品增菌后用EMA-RTQ-PCR进行检测,结果与国标法一致,均为阳性;增菌后的实际样品用EMA-RTQ-PCR进行检测的结果与国标检测结果一致,均为阴性。结论 通过对样品进行前增菌,可以保证检验结果的准确可靠。该方法能够快速、灵敏、准确的检测沙门菌活细胞,具有很好的应用价值。

Abstract:Abstract: Objective To inspect the application value of EMA-RTQ-PCR. Methods We used EMA-RTQ-PCR and GB4789.4-2010 to test artificial simulative samples and actual samples; Results For artificial simulative samples of dead bacterium, the test results of EMA-RTQ-PCR are in accordance with national standard method. For artificial simulative samples of mixed living and dead bacterium, in case of EMA-TRQ-PCR, the results of direct detection are negative, while the results of national standard method are positive. For the mixed enriched sample, the test results of two methods are the same, both are positive. The test results of EMA-RTQ-PCR for actual samples after enrichment are in accordance with national standard method, and both are negative. Conclusion Enrichment can ensure the accuracy of the test results. EMA-RTQ-PCR is reliable through the research, and the method has good application value which can detect living salmonella cells with excellent accuracy and sensitivity.
