摘    要:目的: 探讨和优化红花中羟基红花黄色素A(HSYA)的提取工艺。方法: 应用星点设计-效应面法优化红花中HSYA的提取工艺。采用低温动态法提取红花中的HSYA, 并以提取温度、提取时间和溶剂倍数为考察因素, 以提取率和浓缩液中HSYA含量为评价指标, 采用二次多项式拟合建立考察因素和评价指标之间的数学模型, 根据所建立的数学模型及效应面法预测最佳工艺, 并进行验证。结果: 红花中HSYA最佳提取工艺为:提取温度60 ℃, 加入15倍量水提取2次, 每次66 min。各评价指标预测值与实测值的相对偏差均较小(分别为8.60%, 8.97%), 模型具有良好的预测性。结论: 星点设计-效应面法优选红花中HSYA的提取工艺, 方法简便, 精度较高, 预测性良好。

Abstract:OBJECTIVE To optimize extraction process of hydroxysafflor yellow A from Carthamus tinctorius by central composite design/response surface method. METHODS Central composite design/response surface method was applied to optimize extraction process of hydroxysafflor yellow A from Carthamus tinctorius. Heat soap was used to dynamically extract hydroxysafflor yellow A from Carthamus tinctorius, with extraction temperature, time and solvent ratio as the investigation factors, extraction yield and HSYA content in concentrate as evaluation indexes. Quadratic polynomial fitting was used to construct mathematical model between factors and evaluation indexes. Established mathematical model and response surface method were based to predict the best process and perform validation test.RESULTS Optimal extraction process: extraction temperature 60 ℃, 15 volumes of water, twice extraction, 66 min for each extraction. Relative deviation was small for predicted and determined evaluation index (8.60% and 8.97%, respective). The model had good predictability. CONCLUSION Extraction process of hydroxysafflor yellow A from Carthamus tinctorius optimized by using central composite design/response surface method is simple, has high precision and good predictability.
