IL-1B和IL-1RN基因多态性与非贲门胃癌发病风险的研究

摘    要:摘要:目的 探索IL-1B+3954,IL-1RN-9876,IL-1RN-9739和 IL-1RN-9091基因多态性与非贲门胃癌发病风险之间的关系。方法 采用病例对照研究,收集四川省176名非贲门胃癌患者作为病例组,308例健康者作为对照组。使用自行设计的调查问卷,通过基因组测序检测IL-1B和IL-1RN的基因型。结果 病例组和对照组的男性分别为122例(69.3%)和224例(72.7%),两组的平均年龄分别为(57.7±10.6)和(57.6±11.1)岁,年龄和性别在两组间具有可比性。家族癌症史和饮酒在病例组与对照组之间的差异具有统计学意义(P<0.05)。病例组和对照组携带IL-1B+3954 CC基因位点分别为166例(94.3%)和298(96.8%),IL-1RN-9876 GG分别为141(82.5%)和229(81.5%),IL-1RN-9739 AA分别为145(82.4%)和252(81.8%),IL-1RN-9091 AA分别为60(34.1%)和118(38.3%)。本次研究的4个SNP位点与非贲门癌发病风险之间不存在关联(IL1B+3954 C/T调整OR值1.73,95% CI 0.69~4.30;IL-1RN-9876 G/A调整OR值0.99,95% CI 0.59~1.65;IL-1RN-9739 A/G调整OR值1.02,95% CI 0.62~1.68;IL-1RN-9091 A/C调整OR值1.25,95% CI 0.84~1.86)。此外,对IL-1RN 3个SNP位点进行的单倍体分析尚未发现其是非贲门癌发病的高危因素。结论 四川地区汉族人群IL-1B+3954,IL-1RN-9876,IL-1RN-9739和 IL-1RN-9091基因多态性与非贲门癌发病无关。

Abstract:Abstract: Objective The objective of the study was to explore the correlations between IL-1B+3954, IL-1RN-9876, IL-1RN-9739, and IL-1RN-9091 gene polymorphisms and risk of non-cardia gastric cancer. Methods 176 patients with non-cardia gastric cancer and 308 healthy controls were selected from Sichuan Province, and were divided into a case group and a control group, respectively. All subjects were surveyed by using a self-designed questionnaire, and the genotypes of IL-1B and IL1-RN were determined by genomic DNA sequencing. Results The number of males in the case group and the control group was 122 (69.3%) and 224 (72.7%), respectively, and the average age of the two groups was 57.7±10.6 and 57.6±11.1, respectively; indicating that age and gender were comparable between the two groups. Differences in family cancer history and alcohol consumption were statistically significant between the case group and the control group (P<0.05). The numbers of cases carrying a IL-1B+3954 CC gene locus, a IL-1RN-9876 GG locus, a IL-1RN-9739 AA locus, and a IL-1RN-9091 AA locus were 166 (94.3%), 141 (82.5%), 145 (82.4%), and 60 (34.1%), respectively; and the numbers of controls carrying the same gene loci were 298 (96.8%), 229 (81.5%), 252 (81.8%), and 118 (38.3%), respectively. No significant correlation was observed between any of the four SNPs and risk of non-cardia gastric cancer (IL1B+3954 C/T: adjusted OR 1.73, 95% CI 0.69-4.30; IL-1RN-9876 G/A: adjusted OR 0.99, 95% CI 0.59-1.65; IL-1RN-9739 A/G: adjusted OR 1.02, 95% CI 0.62-1.68; and IL-1RN-9091 A/C: adjusted OR 1.25, 95% CI 0.84-1.86). In addition, haploid analysis of the three IL-1RN SNPs did not indicate that the SNPs were the high-risk factors for non-cardia gastric cancer. Conclusion IL-1B+3954, IL-1RN-9876, IL-1RN-9739, and IL-1RN-9091 gene polymorphisms are not associated with the incidence of non-cardia gastric cancer in the Han population in Sichuan.
