rhBMP-2及rhbFGF对兔骨髓间充质干细胞分化作用长期效应的实验研究

摘    要:目的:检测重组人骨形成蛋白2(rhBMP-2)和重组人碱性成纤维细胞生长因子(rhbFGF)单独、联合及序贯作用对兔骨髓间充质干细胞(BMSC)分化作用的长期效应。方法:采用考马斯亮蓝测定法检测单独、联合及序贯施加rhBMP-2和rhbFGF后兔BMSC在第2、5、7和10天时的蛋白质含量水平,以反映对细胞分化作用的影响,并进行统计学分析。结果:rhBMP-2能在较长时间内明显促进细胞的蛋白质含量增加,而rhbFGF则起抑制作用,并且均呈浓度依赖性。rhBMP-2单独应用与两种生长因子联合应用的效应相近,而且均高于rhbFGF单独应用或两种因子序贯应用的效应。结论:rhBMP-2和rhbFGF的不同应用方式对BMSC分化作用的长期效应不同。

Abstract:Objective:To evaluate the long-term effects of rhBMP-2 and rhbFGF on the differentiation of rabbit bone mesenchymal stem cells by using single agent alone, agents used associatedly, or sequentially. Methods:Coomassie brilliant blue assay was used to evaluate the protein content of bone mesenchymal stem cells so as for reflecting the differentiation of the cells on the 2nd, 5th, 7th, and 10th day by adding rhBMP-2 and rhbFGF with different way. Results:Statistical analysis showed that rhBMP-2 could enhance the protein content of bone mesenchymal stem cells significantly while rhbFGF could inhibit it in long term in a dose-dependent manner. The effects of rhBMP-2 alone was similar to that by using the two growth factors. The effects by two factors were significantly higher than that of using rhbFGF alone and using them sequentially. Conclusion:Different growth factor has different long-term effect on the differentiation of bone mesenchymal stem cells by different ways.
