促红细胞生成素对心肺复苏后大鼠海马神经元NGF、BDNF表达的影响

摘    要:目的观察促红细胞生成素(EPO)对心肺复苏后大鼠海马神经生长因子(NGF)、脑源性神经营养因子(BDNF)表达的影响。方法采用窒息法制作大鼠呼吸心跳骤停模型,然后行心肺复苏术(CPR)。雄性Wistar大鼠60只,随机分3组:手术对照组(A组)、心肺复苏组(B组)、心肺复苏+EPO(C组)。应用免疫组化法观察各组大鼠复苏后12 h海马神经元凋亡细胞、NGF及BDNF表达情况。结果与对照组(A组)相比心肺复苏组(B组)大鼠海马神经元NGF、BDNF表达明显降低(P〈0.05),凋亡细胞明显增加(P〈0.01);心肺复苏+EPO(C组)NGF、BDNF表达明显增加,高于B组和A组(P〈0.01),凋亡细胞明显降低(P〈0.05),低于B组。结论 EPO能促进神经元NGF、BDNF的表达,减少神经细胞凋亡,对缺血、缺氧性神经元损伤有保护作用。

Effect of Erythropoietin on NGF and BDNF in Hippocampal Neurons of Rats after Cardiopulmonary Resuscitation

Abstract:Objective To observe the effects of erythropoietin(EPO)on the hippocampal neurons expression nerve growth factor(NGF) and brain-derived neurotrophic factor(BDNF)in cardiopulmonary resuscitation rat.Methods Cardiac respiratory arrest model were made by apnea method,and followed by CPR treatment.Sixty Wistar rats were randomly divided into three groups: Control group,CPR group and CPR plus erythropoietin group.Expression of apoptotic cell NGF and BDNF in hippocampal neurons were observed by immunohistochemisitry method.Results The expression of NGF and BDNF in hippocampal neurons of rats in CPR group decreased significantly(P〈0.05),and the expression of apoptotic cell increased significantly(P〈0.01);The expression of NGF and BDNF in CPR plus erythropoietin group were much higher than those in control group and CPR group(P〈0.01),the expression of apoptotic cell in CPR plus erythropoietin group decreased significantly(P〈0.05).Conclusion Erythropoietin can increase expression of NGF and BDNF in hippocampal neurons,decrease the number of apoptotic cells and protect neurons from injury of hypoxemia and ischemia.
