五味子甲素与BSA的相互作用

摘    要:目的:在模拟人体生理条件下,研究金属离子对五味子甲素与牛血清白蛋白(BSA)之间结合作用的影响。方法采用紫外吸收光谱法,通过紫外扫描得到五味子甲素与BSA的紫外吸收图谱,利用Lineweavel-Burk双倒数曲线图得出五味子甲素与BSA的结合常数;由摩尔比法确定Me^2+等7种金属离子与五味子甲素结合的摩尔比;分别测定五味子甲素一金属离子-BSA的结合常数。结果五味子甲素与BSA的结合常数为1.141×104L/mol,金属离子与五味子甲素结合的摩尔比为1:1,在Fe^3+、Zn^2+、A1^3+、cu^2+、Mg^2+、Co^2+、Ni^2+存在下,五味子甲素与BSA的结合常数分别为3.041×10^4,1.617×10^4,1.819×10^4,1.781×10^4,1.277×10^3。7.107×10^3,6.238×10^3L/mol。结论Fe^3+、Zn^2+.Al^3+、Cu^2+参与加强了五味子甲素与BSA的结合作用,M^2+、Co^2+Ni^2+的存在住二者结会作用减弱.

Abstract:Objective To study the effect of metalion on the interaction between Deoxyschizandrin and bovine serum albumin(BSA) under the human physiological condition.Methods The UV absorption spectra of Deoxyschizandrin and BSA was obtained by ultraviolet absorption spectroscopy.The binding constant of Deoxyschizandrin-BSA was measured using the Lineweaver-Burk double reciprocal diagram of curve.The proportions of seven metalions binding to Deoxyschizandrin were determined by the method of mole ratio.The binding constants Deoxyschizandrin-BSA were obtained separately.Results The binding constant of Deoxyschizandrin-BSA was K=1.141×104 L/mol.All the mole ratios of Deoxyschizandrin-Men+ was 1∶1 and the binding constants of Deoxyschizandrin-BSA in presence of seven metal ions were 3.041×104,1.617×104,1.819×104,1.781×104,1.277×103,7.107×103,6.238×103 L/mol.Conclusion In presence of Fe3+,Zn2+,Al3+ or Cu2+,the interaction between Deoxyschizandrin and BSA has been strengthened,which has been weakened in presence of other ions.
