摘    要:y-氨基丁酸(GABA)是一种主要的抑制神经递质,其含量的变化与许多神经和精神性疾病有关,本文报道了利用GABA酶偶联细菌荧光酶生物发光系统建立的GABA超微量分析方法。血清标本先经80g/L的三氯乙酸溶液沉淀蛋白,再与双酶工作液混和,由全自动发光光度计记录发光峰值和计算结果。本法的线性范围是0-1000nmol/L。与高效液相色谱法比较,y=0.976,不同含量的3份标本,批内变异分别为11.2%、9.5%和8.3%,批间变异为12.5%。本研究还初步探讨了GABA对帕金森综合征的诊断价值。

Abstract:y-Aminobutyric acid (GABA) is a major inhibitory neumtransimitter found in ceontral norvous system. Abnormalitics in the concentmtion of GABA in many neumlogical and merttal disurders have been reported in the literare.We have developed a sensi- tive , simple and practical method for assaying GABA in serum by uset of was GABAse reations , coupled to a becterial bioluminescence system. The serum pmtcin was precipitated with 80 g/l trichloruacelic acid solution , mixed with two enzyme working solution , and the peaka of Iuminescence were recorded . The linearty was found to be from 0 to 1000 nmol/s of GABA . In accuracy atudies , the results of biolutainescence method were compared with those of high performance performance liquid chromatography(HPLC)and showed a correlation coeffi- cient of 0 . 976 . The CV values in the three sentm samples containinp low , medium and high levels of GABA were 11. 2%, 9.5%, and 8.3% for within batches , and thc CV value was 12. 5% for between hatches. The study suggests that the determination of GABA in serum might be a potential biochemical marker of Parkinsun disease.
