大黄配方颗粒的UHPLC指纹图谱研究

摘    要:目的:建立大黄配方颗粒的UHPLC指纹图谱,为配方颗粒的快速质量评价提供方法。方法:采用超高效液相色谱法,以Agilent pomshell 120EC C18柱(4.6mm×100mm,2.7μm)为色谱柱;以甲醇:0.4%冰醋酸水溶液为流动相梯度洗脱;检测波长254nm;流速0.8mL/min;柱温25℃。结果:建立了大黄配方颗粒的UHPLC指纹图谱,确定了17个共有峰,各大黄配方颗粒样品指纹图谱与对照指纹图谱相似度均在0.9以上,可以用于大黄配方颗粒的快速定性鉴别。结论:该方法简单、准确、快速、重复性好,能有效地对大黄配方颗粒的质量进行评价。

Study on UHPLC fingerprint analysis of Rhei Radix et Rhizoma Formula Granules
Abstract:Objective:To establish a method of fingerprint analysis on Rhei Radix et Rhizoma Formula Granules by UHPLC and provide a scientific method for rapid quality evaluation of Rhei Radix et Rhizoma Formula Granules. Methods:The chromatographic fingerprints were obtained by the Agilent poroshell 120 EC C18 column(4.6mm×100mm, 2.7μm) with the gradient elution solvent system composed of methanol - 0.4%glacial acetic acid. The detective wavelength was set at 254nm;the flow rate was 0.8 mL/min and the column temperature was maintained at 25℃.Resuhs: 17 co-possessing peaks were selected as the fingerprint peaks of Rhei Radix et Rhizoma Formula Granules and good similarities with correlation coefficients higher than 0.9 were found in fingerprints between the different samples of Rhei Radix et Rhizoma Formula Granules and the standard fingerprint. Conclusions: The method with good reproducibility is simple, rapid and accurate, it can be used as a quality control method for Rhei Radix et Rhizoma Formula Granules.
