百合知母汤抗抑郁的药效研究

摘    要:目的:观察百合提取物(Lily extracted substances,LES)、知母提取物(Zhimu extracted substances,ZES)、百合知母汤提取物(Lily—Zhimu extracted substances,L—ZES)对慢性不可预见性温和刺激(Chronic unpredictable mild stress,CUMS)配合孤养所致大鼠抑郁模型的影响。方法:60只雄性SD大鼠随机分为正常组、模型组、百合提取物组、知母提取物组、百合知母汤提取物组、阳性药氟西汀组。采用CUMS配合孤养诱发抑郁模型。连续灌胃给药6周,观察大鼠体重变化,运用旷场实验、糖水偏爱实验评定大鼠的行为学特征。以酶联免疫吸附法(ELISA)检测血清多巴胺(DA)、促肾上腺皮质激素(ACTH)的含量。结果:与正常组相比,模型组大鼠的体重增加缓慢、行为学特征下降明显,血清DA含量下降、ACTH含量升高,差异有统计学意义(P〈0.01),说明CUMS配合孤养可致大鼠出现抑郁行为。与模型组比较,百合提取物组、知母提取物组及百合知母汤提取物组、阳性药氟西汀(FHP)组能显著对抗抑郁刺激,大鼠的体重增加、行为学特征改善,DA的含量提高ACTH的含量降低。结论:百合提取物、知母提取物、百合知母汤提取物有很好的抗抑郁作用,其机制可能与调节抑郁症模型大鼠体内神经递质紊乱及调节HPA轴的功能、抑制大鼠HPA轴亢进有关。

Antidepressant Efficacy Studies on Lily Zhimu Decoction

Abstract:Objective:To observe the antidepressant effect of LES;ZES;L-ZES on the model of the depressive rats that are induced by the CUMS and separate feeding method. Methods 60 male SD rats were randomLy divided into normal group, model group, LES group, ZES group, L-ZES group, positive drug fluoxetine (FHP)group. Use CUMS and separate feeding induced model of the depressive rats, All the rats are continuously gavaged for 6 weeks , and their data of changing weight were taken.And the Open Field Test and the Preference for Sucrose Test were used to assessed behavioral characteristics of the rats. The content of serum DA and ACTH were detected by using ELISA.Results Compared with the normal group,the model group gained weight slower and their behavioral characteristics decreased significantly. The DA content in serum decreased and the ACTH increased .The difference was statistically significant (P〈0.01).It indicates that CUMS and isolated raising can induce depression phenomenon in rats .When it is compared with model group,the LES group; the ZES group;the L-ZES group and the FHP group could antagonize depression stimulation .The rats gained weight and improved the behavioral characteristics,and the content of DA increased and the ACTH's decreased. Conclusion LES, ZES, L-ZES have a positive effect on antidepressant, and the mechanism be related to adjustment in vivo neurotransmitter disorders and regulation HPA axis hyperactivity.
