大鼠哮喘模型中嗜酸性粒细胞和IL-10改变研究

摘    要:目的探讨大鼠哮喘模型中白细胞介素-10(IL-10)、嗜酸性粒细胞(EOS)的变化及其在哮喘发病机制中的作用。方法30只SD大鼠随机分为正常对照组、3周哮喘组和4周哮喘组,用ELISA法测定各组大鼠外周血清IL-10,并计数外周血和肺泡灌洗液(BALF)中EOS。结果各哮喘组IL-10显著低于对照组(P均<0.05),且4周哮喘组IL-10显著高于3周哮喘组(P均<0.05);外周血EOS计数各哮喘组显著高于正常对照组(P均<0.05);在BALF中,各哮喘组显著高于正常对照组(P均<0.01),但2个哮喘组间无显著性差异。血清IL-10浓度与外周血和BALF中EOS绝对计数无显著相关性。结论大鼠哮喘模型中有血清IL-10下降现象,在4周后有升高趋势,但仍低于正常对照组,其升高趋势与EOS计数无线性相关,IL-10升高趋势与气道重塑可能相关。

Investigate of changes of interleukin - 10 and Eosinophil in rat asthma model
Abstract:Objective It is to approach the changes of interleukin-10(IL-10) and Eosinophil(EOS) in the tats asthma model,and approach the role of them in the pathogenesis of asthmatic attack.Methods Thirty SD rats were randomly divided into 3 groups: normal control group,asthma for 3 weeks group,asthma for 4 weeks group.The serum levels of IL-10 were determined by ELISA method.And the counts of EOS in blood and BALF were measured.Results The serum levels of IL-10 in two asthma groups decreased significantly compared with the control group respectively(P all<0.01).Asthma for 4 weeks group was significantly higher than those in asthma for 3 weeks groups(P all <0.01).The counts of EOS in blood in two asthma groups were increased markedly compared with that in normal control group(P<0.01);in BALF,the counts of EOS in two asthma groups were increased markedly compared with that in normal control group(P all <0.01),and there was not significantly in the both asthma groups. There were not significant correlation among serum levels of IL-10 and number of EOS in blood and BALF.Conclusion The serum levels of IL-10 are reduced in rat asthma models,and may increase after 4 weeks,but they are remain reduced compare with the normal control group.There was no correlation in the levels of IL-10 and the counts of EOS.The increased expression of serum levels of IL-10 suggested that it play a significant role in airway remodeling of asthma.
