多组分反应及其在新药研究中的应用

摘    要:本文对多组分反应的发展进行了回顾,简述了其在结构多样性和复杂性的化合物和化合物库合成上的优势,以及在新药研究中新先导物的发现和结构优化上的重要应用价值,简要介绍了几种重要的多组分反应和探索新多组分反应的方法。

Abstract:The organic reactions that involve three or more different starting materials as chemical structure inputs and give rise to one final product in a one-pot procedure are called multi-component reactions(MCR).Owing to their advantages in generation of small organic molecules with structural diversity and complexity,MCR have been a resurgence of interest in academy and industry of chemistry since 1990s.The history of MCR,some important reaction types,such as the Ugi reaction and the Passerini reaction,and some representative examples applied to drug discovery are introduced in this review.Three methods to the discovery of new MCR are combinatorial reaction finding,rational reaction design and Union of MCR.
