NF2基因对细胞增殖和细胞周期的影响

摘    要:目的观察Ⅱ型神经纤维瘤(neurofibromatosisⅡ,NF2)基因对细胞增殖和细胞周期的影响。方法采用脂质体介导法将携带野生型NF2的真核表达载体导入大鼠神经鞘瘤RT4细胞,质粒转染成功后实验细胞分为pEGFP-N1组、pEGFP-NF2组和对照组(未转染质粒组),以Western blot分析目的基因的表达,并采用MTT法和流式细胞仪检测细胞增殖和细胞周期。结果pEGFP-NF2组细胞merlin蛋白出现明显高表达。pEGFP-NF2组细胞生长的抑制率高达37.7%,与对照组相比较,差异具有统计学意义(P〈0.05);而pEGFP-N1组细胞生长的抑制率与对照组比较,差异无统计学意义(P〉0.05)。流式细胞仪检测显示NF2可以阻断细胞周期,使其处于G1-G0期。结论野生型NF2可抑制大鼠神经鞘瘤RT4细胞的增殖,阻断细胞周期,使其处于G1-G0期。

Effect of the transfected NF2 gene on cell cycle and proliferation of rat schwannoma cell line

Abstract:Objective To investigate the effect of the neurofibromatosis Ⅱ (NF2) gene on cell cycle and proliferation in rat schwannoma cell line. Methods The eukaryotic expression vector pEGFP-NF2 containing NF2 cDNA was transfected into RT4 cells of the rats by lipofectamine and then these cells were divided into pEGFP-N1 group, pEGFP-NF2 group and the control group (cells without transfection). The NF2 expression in the transfected RT4 cells was detected by Western-blotting. The cycle, proliferation and apoptosis of the RT4 cells were determined by the MTT and flow cytometry. Results High expression of merlin protein was found in the transfected RT4 cells of the pEGFP-NF2 group. The proliferation of pEGFP-NF2 cells were inhibited by 37,7% (P 〈 0.05 vs. the control group), while pEGFP-N1 cells showed no significant change in proliferation (P 〉 0.05 vs. the control group), The cell cycle from G1 to G0 of pEGFP-NF2 cells was inhibited. Conclusion Wide type NF2 gene might suppress the growth and induces apoptosis of rat schwannoma cell line RT4 cells.
