姜黄素对大鼠腹膜纤维化的作用及其对BMP-7、TGF-β1,表达的影响

摘    要:目的探讨姜黄素对腹膜纤维化大鼠腹膜的作用及对腹膜骨形态发生蛋白-7(BMP-7)、转化生长因子-β1(TGF-β1)表达的影响。方法将36只雄性SD大鼠随机分为对照组、模型组、姜黄素组各12只。28d后,通过腹膜平衡试验及HE染色、Masson染色对大鼠腹膜进行组织结构和功能检测。利用免疫组化对BMP-7和TGF-β1,在腹膜的表达进行检测。结果与对照组和姜黄素组相比,模型组大鼠超滤量和初始腹膜透析液葡萄糖浓度(D0)与透析液葡萄糖浓度(D1)比值(D1/D0)明显减少,而透析液尿素浓度与血浆尿素浓度比值(Durea/Purea)明显增加(P〈0.05)。模型组的腹膜组织厚度较对照组和姜黄素组明显增加(P〈0.05)。免疫组化可见与对照组相比,模型组TGF-β1,表达明显增加(P〈0.05);而姜黄素组较模型组TGF--β1,表达明显减少(P〈0.05)。BMP-7在对照组中高度表达,而在模型组中表达明显减少(P〈0.05),姜黄素组BMP-7表达较模型组明显增加(P〈0.05)。结论姜黄素对腹膜纤维化有明显疗效,其机制可能是通过上调BMP-7的表达,拮抗TGF-β1的作用,从而抑制腹膜纤维化。

Therapeutic effect of curcumin on peritoneal fibrosis and its influence on the expression of BMP-7 and TGF-β1 in rats with peritoneal fibrosis

Abstract:Objective To explore the therapeutic effects of curcumin on peritoneal fibrosis and its influence on the expression of bone morphorgenetic protein-7 (BMP-7) and transforming growth factor-β1 (TGF-β1) in rats with peritoneal fibrosis. Method Thirty-six SD rats were randomly divided into 3 groups., control group, model group and curcumin-treated group. Animals were sacrificed after 28 days. The structure and function of the peritoneum were examined by peritoneal equilibration test, HE staining and Masson staining. The expression of BMP-7 and TGF-β1 was detected by immunohistochemistry straining. Result Compared with the control group and curcumin-treated group, the D1 / D0 ratio and net ultrafiltration volume in model group were significantly reduced (P〈0. 05), while the Durea/Purea ratio in model group was significantly increased (P〈0. 05). The thickness of the peritoneal membrane in model group was significantly greater than that in control group and curcumin-treated group (P〈0. 05). Immunohistochemistry staining of peritoneal membrane showed that the expression of BMP-7 in model group was obviously lower than in curcumin-treated group, while the expression of TGF -β1 was obviously higher than in curcumin-treated group. Conclusion Curcumin can effectively inhibit peritoneal fibrosis possibly by up-regulating the expression of BMP-7, antagonizing the effects of TGF -β1.
