摘 要: | 目的:研究血链球菌细菌素对单独及混合培养的白色念珠菌(C.a)及热带念珠菌(c.t)细胞表面疏水性的影响。研究血链球菌细菌素对单独及混合培养的白色念珠菌(C.a)及热带念珠菌(C.t)细胞膜钙离子通透性的影响。方法:复苏并传代血链球菌标准株ATCCl0556,通过低温高速离心,细胞破碎等方法提取血链素,使其作用于1×10。du/mL单独及混合培养的C.a及C.t,于37℃,80r/min振荡培养。1)采用二倍稀释法,测定血链素对C.a及C.t最小抑制浓度(MIC);2)运用微生物黏着碳氢化合物的方法[1],用紫外分光光度计测A值,计算C.a及c.t的疏水率;3)分别向单独及混合培养的C.a及C.t菌悬液中装载钙离子荧光探针(Fluo-3AM),利用激光共聚焦显微镜观察血链素作用后的C.a及C.t细胞内钙离子浓度的变化。结果:血链素对单独培养的C.a及C.t的MIC为0.25g/L;血链素对等量混合培养的C.a及C.t的MIC为0.5g/L。2)血链素作用后,单独及混合培养的C.a及C.t细胞表面疏水率均明显降低,与对照组相比具有显著差异(P〈0.05)。在血链素的作用下,单独及混合培养的C.a及C.t的细胞内钙离子浓度均明显降低,与对照组相比具有统计学意义(P〈0.05)。结论:血链素可以导致白色念珠菌及热带念珠菌细胞表面疏水性降低。血链素可以导致白色念珠菌及热带念珠菌细胞膜钙离子通透性增强。

Abstract: | Objective: To study the cell surface hydrophobicity and membrane permeability of Streptococcus san- guis bacteriocin on Candida albicans(C, a) and Candida tropicalis (C. t). Methods: Recovery and extend standard strains ATCC10556,Streptococcus sanguis bacteriocin was extracted by low--temperature high--speed centrifuga- tion, ultrasonic disruption, and then was respectively applied to separate and mixed cultured C. a and C. t under 37℃ 80r/min oscillating culture. 1. Used Adopt double dilution method to determine the MIC of C. a and C. t in un- der the action of Streptococcus sanguis bacteriocin. 2. Using the method of microbial gelling hydrocarbons, measuring absorbance value with Ultraviolet spectrophotometer and calculating hrdrophobic. 3. Adding the Fluo--3 AM to C. a and C. t and observing concentration of calcium ion into cell with Laser scanning confocal microscope. Results: The MIC of separate C. a and C. t is 0.25g/L, The MIC of mixed C. a and C. t is 0.5g/L. Under the influence of Sanguis Bacteriocin hrdrophobic of C. a and C. t reducing significantly, compared with the control group have significant difference. 3. Under the influence of Sanguis Bacteriocin concentration of calcium ion into cell of C. a and C. t de- creased significantly,compared with the control group have significant difference. Conclusion: Streptococcus sanguis bacteriocin could significantly inhibit cell surface hrdrophobicity of separate and mixed cultured C. a and C. t. Strep- tococcus sanguis baeteriocin enhanced the calcium ion permeability of cell membrane of C. a and C. t. |
