山茱萸不同极性提取物降糖作用的研究

摘    要:目的:对山茱萸不同极性的提取物进行初步药理学评价,探讨其对正常小鼠、四氧嘧啶糖尿病模型小鼠血糖、血脂及胰岛素的影响。方法:(1)将山茱萸的水提取物、醇提取物、乙酸乙酯提取物、石油醚提取物(浓度皆为1g生药材/mL溶液)以7.5g生药材.kg-1对正常小鼠灌胃,连续7天,观察其降血糖作用。(2)通过灌胃四氧嘧啶(Alloxan)50mg.kg-1×3天分次注射造模建立的小鼠糖尿病模型,以相同剂量的山茱萸不同极性提取物连续7天灌胃,观察其降血糖作用及对胆固醇、甘油三酯和胰岛素水平的影响。结果:山茱萸不同极性提取物对正常小鼠的血糖水平无明显影响。山茱萸醇提取物、乙酸乙酯提取物组与模型组相比,可显著降低四氧嘧啶糖尿病模型小鼠的血糖值(P<0.05),其中山茱萸醇提取物可显著提高血清胰岛素水平(P<0.05);山茱萸乙酸乙酯提取物可显著降低糖尿病模型小鼠血清中甘油三酯含量(P<0.01),山茱萸醇提取物对甘油三酯有降低趋势。结论:山茱萸醇提取物、乙酸乙酯提取物对糖尿病模型动物具有较好的降血糖和降血脂作用。

Abstract:Objective:To investigate the antidiabetic effects of different extraction from fructus corni on animal models of diabetes mellitus. Methods:The normal mice received a daily intra-gastric dosage of 7. 5g·kg -1 with water-soluble extraction,alcohol-soluble extraction,acetate-soluble extraction and ligarine-soluble extraction of fructus corni for 7 days,then the fasting serum glucose (FSG) was detected. The alloxan-induced diabetes mice by iv 50mg·kg -1 × 3d,were given the same dose of extraction of fructus corni for 7 days,the FSG,cholesterol,triglyceride and fasting insulin were detected. Results:The glucose had no significant change in normal mice with different extraction of fructus corni. The glucose in diabetes mice was significantly decreased (P 0. 05) compared with model by alcohol-soluble extraction and acetate-soluble extraction of fructus corni,The fasting insulin level elevated(P 0. 05) in alloxan-induced diabetes mice with alcohol-soluble extraction,while triglyceride level also decreased significantly with acetate-soluble extraction. Conclusion:The alcohol-soluble extraction and acetate-soluble extraction of fructus corni have remarkable hypoglycemic effect on diabetes animal models.
