Abstract:Objective:To explore the influence of hypoxia on proliferation and differentiation of neural stem cells(NSCs) of hippocampus in neonate rats.Methods:Eighteen neonate rats(group Q1,6 rats,1 day after birth;group Q7,6 rats,7 days after birth;group Q14,6 rats,7 days after birth) were exposed to hypoxia status for 2 hours.Then hippocampi of each group were put in the serum-free medium for monoclonal culture or subcultivation.Then,monocloned cells were induced to be differentiated by fetal bovine serum for 3 days,and then nestin positive cells were identified by immunofluorescence.Neuron specific enolase(NSE) positive or glial fibrillary acidic protein(GFAP) positive cells of 3G subcultivated NSCs were calculated.Results:All monoclonal cells were nestin positive;while only parts of the subcultivated cells were NSE positive or GFAP positive.It took 7d,11d and 40d for the monoclonal cells of group Q1,Q7 and Q14 to develop into a diameter of 200μm.The percentage of NSE positive cells was significant higher in Q1 than that in Q14.Conclusions:Hypoxia pretreatment can promote proliferation of hippocampus NSCs and increase the percentage of NSE positive cells more obviously Q1 rats than that in Q 14 rats.
