长链非编码RNA PVT1对人脑胶质瘤U251细胞增殖能力的影响

摘    要:目的 探讨长链非编码RNA PVT1(lncRNA-PVT1)对人脑胶质瘤U251细胞增殖能力的影响。方法 体外培养胶质瘤细胞系U251细胞,分别转染PVT1 siRNA(PVT1组)和negative control siRNA(对照组)。PCR法检测lncRNA-PVT1及C-MYC mRNA表达水平,CCK-8法检测细胞增殖能力,Western blot检测C-MYC蛋白表达水平。结果 PVT1组lncRNA-PVT1 mRNA表达水平(0.27±0.08)较对照组(1.0±0.30)明显降低(P<0.05);PVT1组C-MYC mRNA表达水平(0.87±0.19)与对照组(1.0±0.15)无明显差异(P>0.05)。转染siRNA后1、2、3 d,PVT1组U251细胞活力较对照组均明显降低(P<0.05)。PTV1组C-MYC蛋白表达水平(0.29±0.12)较对照组(0.85±0.27)明显降低(P<0.05)。结论 降低lncRNA-PVT1表达水平能够抑制U251细胞增殖,其机制可能与lncRNA-PVT1够影响C-MYC蛋白的表达水平有关。

Abstract:Objective To explore the effect of long non-coding RNA PVT1 (lncRNA-PVT1) on the proliferation of glioma U251 cells. Methods The cultured glioma U251 cells were divided into 2 groups, i.e. experimental group in which PVT1 small interfere RNA (SiRNA) were tansfected into U251 cells and control group, in which negative control SiRNA was transfected into U251 cells. The expressions of lncRNA-PVT1 and C-MYC mRNA were detected by real-time quantitative PCR (RT-qPCR) in both the groups, in which the expressions of C-MYC protein were detected by Western blot. The proliferation of the U251 cells was determined by Cell Counting Kit-8 (CCK8) assay in two groups. Results The expression of lncRNA-PVT1 mRNA (0.27±0.08) was significantly lower in the experimental group than that (1.0±0.30) in the control group (P<0.05), but there is no significant difference in the expression of C-MYC mRNA between the two groups (P>0.05). The expression of C-MYC protein was significantly higher in control group than that in the experimental group (P<0.05). The cells viability was significantly lower in the experimental group than that in the control group 1, 2 and 3 days after the transfection (P<0.05). Conclusions Down-regulating the expression of lncRNA-PVT1 can decrease the expression of C-MYC protein in U251 cells and inhibit the proliferation of U251 cells.
