益肾降浊汤对脑缺血再灌注大鼠海马NE、5-HT含量的影响

摘    要:目的 观察益肾降浊汤对脑缺血再灌注大鼠海马NE、5 HT含量的影响。方法 阻断大鼠双侧颈总动脉进行缺血再灌注 ,于术后d 1、d 7、d 15用高压液相色谱电化学检测法测定海马NE、5 HT的含量。结果 模型大鼠在术后d 1、d 7、d 15海马NE含量分别是 :36 4 2 5± 6 6 4 7,349 76± 5 9 38,(344 5 9± 70 31) μg·g-1,5 HT含量分别是 :6 4 6 72± 83 33,6 2 9 11± 90 6 4 ,(5 96 6 8± 99 4 7) μg·g-1,均较正常组明显降低 (P <0 0 1) ,且随着时间的延长逐渐下降。而中药组大鼠在相同日期海马NE含量分别是 4 17 91± 6 8 2 2 ,4 16 37± 4 8 6 5 ,(42 0 2 9± 5 1 4 5 ) μg·g-1,5 HT含量分别是 :731 31± 5 7 2 6 ,74 6 4 5± 77 77,(770 2 4±5 8 4 1) μg·g-1,均明显高于模型组 (P <0 0 5 )。结论 NE和 5 HT参与了脑缺血再灌注后的神经元损伤 ,益肾降浊汤可升高脑缺血再灌注后的海马NE、5 HT含量 ,保护神经元。

Abstract:AIM To observe the effect of Yishenjiangzhuo decoction on the NE,5-HT contents of hippocampus in cerebral ischemia reperfusion rats. METHODS High pressure lipuid chromatography-electrochemical process was used to measure the NE and 5-HT contents on the d 1, d 7 and d 15 after cerebral ischemia reperfusion by common carotid artery occlusion. RESULTS The NE contents of hippocampus were respectively 364.25±66.47, 349.76±59.38, (344.59±70.31) μg·g -1 and the 5-HT contents were respectively 646.72±83.33,629.11±90.64,(596.68±99.47) μg·g -1 on the d 1, d 7 and d 15 in the model group, which significantly decreased compared with the control group( P <0.01). The NE and 5-HT contents decreases become more and more severely over time. While in the Yishenjiangzhuo decoction group the NE contents of hippocampus were respectively 417.91±68.22,416.37±48.65,(420.29±51.45) μg·g -1 and the 5-HT contents were respectively 731.31±57.26,746.45±77.77,(770.24±58.41) μg·g -1 on the d 1, d 7 and d 15, while significantly increased compared with the model group on the same experimental day( P <0.05). CONCLUSION The NE and 5-HT play a major role in neurons damage of cerebral ischemia reperfusion, Yishenjiangzhuo decoction might increase the NE and the 5-HT contents of hippocampus in cerebral ischemia reperfusion rats and preserve neurons.
