丹参素对糖尿病脑梗塞大鼠t-PA、PAI-I含量及mRNA表达的影响

摘    要:目的观察丹参素对糖尿病脑梗塞大鼠脑组织纤溶酶原激活物及其抑制剂mRNA表达,及对血浆纤溶功能的影响,探讨其可能作用机制。方法糖尿病大鼠丹参素治疗一周后,应用自体血栓注入大鼠脑动脉制成糖尿病脑梗塞模型。ELISA方法检测血浆中t-PA、PAI-I含量,RT-PCR方法测定脑组织中t-PA与PAI-I,u-PA与NSP的mRNA表达。结果丹参素治疗组促纤溶因子t-PA含量及mRNA表达升高、抑制因子PAI-I含量及mRNA表达降低。作用强度随丹参素浓度的增大而增强。u-PAmRNA、NSPmRNA的表达无显著性变化。结论丹参素可显著改善糖尿病脑梗塞大鼠血浆纤溶功能,通过促进t-PA含量及其mRNA表达、抑制PAI-I含量及其mR-NA表达,而提高鼠脑纤溶功能。

Abstract:Objective To observe the influence of Danshensu pretreatment on the expression of mRNA of plasminogen activator and plasminogen activator inhibitor and the function of plasma fibrin dissolve in diabetic rats with cerebral infarction. To study the role of danshensu in prevention and treatment of diabetic cerebral infarction and its possible mechanism. Methods The diabetic rats were injected Danshensu by tail vein. After one week, cerebral infarcted models were made. PAI-I and t-PA in the plasma were detected with ELISA. The expression of mRNA of t-PA and PAI-I, u-PA and NSP, were detected with RT-PCR. Results Danshensu pretreatment can improve plasma fibrin dissolve of diabetic rats with cerebral infarction.The expression of t-PA mRNA increased and PAI mRNA decreased, And the effect was more and more enhanced when the content of danshensu increased, nevertheless there was no significantly effective on the expression of u-PA and NSP mRNA. Conclusions Danshensu pretreatment can improve the function of plasma fibrin dissolve significantly, and it in brain by increasing t-PAmRNA and decreasing PAImRNA in diabetic rats with cerebral infarction. The effect might relate to the function, which protecting endothelial cells and invigorating the circulation of blood, so it might react on the prevention and treatment of diabetic cerebral infarction.
